Background of the Invention
This invention relates to methods of delivering nucleic acids into cells.
Gene therapy is a highly promising technique for treatment of hereditary diseases, e.g., cystic fibrosis. Gene therapy can also be used when expression of gene products from genes which are not naturally found in the host cells is desired, for example, from genes encoding cytotoxic proteins targeted for expression in cancer cells.
Gene therapy can fall into several categories. It is sometimes desirable to replace a defective gene for the entire lifespan of a mammal, as in the case of an inherited disease such as cystic fibrosis, phenylketonuria, or severe combined immunodeficiency disease (SCID). In other cases, one may wish to treat a mammal with a gene that will express a therapeutic polypeptide for a limited amount of time, e.g., during an infection. Nucleic acids in the form of antisense oligonucleotides or ribozymes are also used therapeutically. Moreover, polypeptides encoded by nucleic acids can be effective stimulators of the immune response in mammals.
Various techniques have been used for introducing genes into cells, including infection with viral vectors, biolistic transfer, injection of "naked" DNA (US Patent No. 5,580,859), and delivery via liposomes or polymeric particles.
In WO 9524929, encapsulated supercoiled circular DNA plasmids are manufactured in a process comprising solvent removal.

Hintergrund der Erfindung
Diese Erfindung bezieht sich auf Verfahren zum Liefern von Nukleinsäuren in Zellen.
Gentherapie ist ein sehr viel versprechendes Verfahren zur Behandlung von Erbkrankheiten, z. B. zystischer Fibrose. Gentherapie kann auch verwendet werden, wenn die Expression der Genprodukte von Genen gewünscht wird, welche natürlicherweise nicht in den Wirtszellen gefunden werden, zum Beispiel von zytotoxische Proteine kodierenden Genen, die auf eine Expression in Krebszellen zielen.
Gentherapie kann in verschiedene Kategorien fallen. In einigen Fällen ist es wünschenswert, ein defektes Gen für die gesamte Lebensspanne eines Säugers zu ersetzen, wie in dem Fall einer Erbkrankheit wie zum Beispiel zystische Fibrose, Phenylketonurie oder schwerer kombinierter Immundefekt (SCID). In anderen Fällen kann es erwünscht sein, einen Säuger mit einem Gen zu behandeln, das ein therapeutisches Polypeptid für eine begrenzte Zeitdauer exprimieren wird, z. B. während einer Infektion. Nukleinsäuren in Form von Antisense-Oligonukleotiden oder Ribozymen werden auch für therapeutische Zwecke genutzt. Außerdem können von Nukleinsäuren kodierte Polypeptide effektive Stimulatoren der Immunantwort in Säugern sein.
Verschiedene Verfahren wurden für das Einbringen von Genen in Zellen verwendet, einschließlich der Infektion mit viralen Vektoren, der biolistische Transfer, der Injektion "nackter" DNA (US-Patent-Nr. 5,580,859) und der Lieferung über Liposomen oder Polymerpartikel.
In WO 9524929 werden eingekapselte zirkuläre DNA-Plasmide in Supercoil-Form in einen Verfahren hergestellt, welches die Entfernung von Lösungsmittel umfasst.
