Establishment of myogenic primary culture

Контекст: In addition to studies of fat tissue function and alterations, the SICA-HF consortium aims to identifiy and characterize
functional, ultrastructural and molecular changes in skeletal muscle tissue in CHF that could be considered as early
markers of worsening clinical evolution of the disease (WP08). Furthermore, we aim to gain insight into the cross-talk
between muscle, fat and the immune system in these pathological conditions. For this purpose, we plan to harvest
skeletal muscle biopsies We plan to establish myogenic primary cultures in order to evaluate several parameters of
proliferation and myogenic differentiation, as size of myotubes, correct expression of the muscle specific regulatory
factors, cell cycle regulators and terminal differentiation markers. Since skeletal muscle homeostasis is maintained
by the regenerative capacity of muscle stem cells (satellite cells), the ability to proliferate, differentiate and fuse
leading to new muscle formation of satellite cells isolated from biopsies of CHF patients will be also analyzed.
