After initial titration studies to identify optimum chimpanzee cell binding concentrations with CalTag and IDEC-114 reagents, we obtained peripheral blood samples from three normal healthy chimpanzees. A control blood sample (0.1 mL) from each animal was stained with the three color markers. Concurrently unlabeled IDEC-114 was added or "spiked" in a final concentration of 30 ug/mL to a second aliquot of blood to simulate in vivo treatment, then incubated for one hour and stained with the same reagents. The B lymphocyte population was subsequently selected by gating on the PerCP labeled CD20 population and gating out all other events. The number of cells located in the RPE single stained populations and RPE/FITC double stained populations from control and "spiked" test samples were compared.
