I'm an over-the-phone interpreter, and I am always on a 3-way call with an English-speaking person and a Chinese-speaking person. When the English person says "I will need to put you on hold for 2 minutes," I can't find a very accurate term in Chinese to say to the Chinese person. I always say 请稍等两分钟 but the Chinese always keep on talking instead of knowing that they are already on hold and the English person will not be back in 2 minutes. How can I interpret the term accurately so the Chinese person know I'm not only asking them to wait for 2 minutes, but they are actually also on hold?

I think your own 请稍等两分钟 is still the best so far. No matter how you interpret it, some people would still keep talking. It is not because they ddin't understand you, instead, they are just trying to confirm you and the talker are still there ( to make sure the line is not disconnected).
