Elle a été initialement sélectionnée par sa capacité de fixation à la glycoprotéine d’enveloppe gp120 du VIH1. Ce n’est qu’en 2000 que les études de l’équipe de Geijtenbeek et van Kooyk ont révélé l’importance fonctionnelle de cette molécule pour les CD. Cette équipe a montré qu’un ligand cellulaire de DC-SIGN était la molécule ICAM3 (d’où son nom DC-specific ICAM-3 grabbing non integrin) [10]. La même équipe a ensuite montré que DC-SIGN se liait également à ICAM2 [11].
