You take a circular piece of DNA (PLasmid) that is cut at one point, you add two different pieces of linear DNA and ligate the whole lot so you get the three pieces linked into a circle ("ligated" at three different points).

agree michelebe: but the french term "ligature" rather than "ligation" is mentioned in different genetic glossaries... wouldn't it be more suitable?
