"For the molecular marker analysis, DNA was isolated from 50 mg of leaves with EZNA HP Plant DNA Kit according to the manufacturer protocol (Omega Bio-Tek). Polymorphisms of 14 microsatellite loci were assessed in a maximum of 95 plants from the living collection of Diputación Provincial de Pontevedra. The reactions were performed in 25 μL total volume containing 50 ng of genomic DNA. Cycling parameters were as follows: 5 min at 95°C, 25 cycles for 1 min at 95°C, ***45 s*** of hybridization with corresponding Tm (Table 2), 1 min at 72°C, 20 cycles for 1 min at 95°C, ***45 s*** of hybridization at 55°C Tm, 1 min at 72°C and a final step of 60 min at 72°C."
