Az ilyen gátlás inkább azt a tényt tükrözi, hogy az így szabályozott enzimek allosztérikusak: az enzim nem csupán a szubsztrátkötésre alkalmas katalitikus hellyel rendelkezik, hanem még egy vagy több más, olyan hellyel is, ahol kicsiny, a szubsztráttól eltérõ szerkezetû, szabályozó molekula, effektor vagy ligand megkötése válik lehetségessé (allosztéria = más alakú). Az effektor megfelelõ helyen kötõdve az enzim konformációváltozását idézi elõ, és ezáltal a katalitikus hely szubsztrátra vonatkoztatott affinitása csökken (allosztérikus gátlás). Van azonban olyan eset is, amikor az effektor kötés hatására éppen megnõ az aktivitás (allosztérikus aktíválás).
