Upon transfection of the host cell, the repressor is expressed in the cell and binds to the repressor binding site of the selection component upstream to the selection gene to thereby suppress expression of the selection gene. Once tetracycline is added to the system, it binds to the tetracycline repressor and immediately detaches from the binding site on the DNA which, in turn, leads to the expression of the selection gene. Consequendy, expression of the selection gene is dependent on the addition of tetracycline and, therefore, expression of the selection gene is ***rendered induciblе***.
