"HPLC analyses of mifepristone and HRP 2000 in serum were carried out by adaptation of the method previously developed for the direct analysis of tamoxifen in serum, by incorporating on-line extraction using an ISRP-cartridge. Samples were collected once pre-treatment, daily through two days post-treatment (GD 23-28), then every other day from GD 30 to 40. In brief, samples collected for mifepristone analysis were diluted 1:10 with 0.01 M phosphate buffer at pH 6.0. HRP 2000 samples were diluted 1:l with 0.01 M phosphate buffer at pH 8.0. Both antiprogestin solutions were centrifuged at 3,000 rpm for 15 min to remove any precipitated protein. The mifepristone supernatant **was loaded to overfill** in a 1 ml injection loop in a xxx HPLC injector whereas 200-500 µl of the HRP 2000 supernatant was loaded. "
