Term ContextMetabotropic glutamate receptors play a role in a wide variety of neuronal functions. Exaggerated mGlu5 signaling may account for the cognitive impairment and other clinical symptoms associated with FXS. Preclinical data supports this mGlu5 theory and suggests there is a ‘rescue’ of several phenotypes of the Fragile X disease model based upon a decrease in mGlu5 receptor functioning either by administration of an mGlu5 antagonist or a decrease in the abundance of receptors. Studies with the mGlu5 antagonists MPEP (2-Methyl-6-(phenylethynyl)-pyridine) [10] and fenobam [17] have demonstrated that acute inhibition of the mGlu5 receptor in adult FMR1 knockout mice leads to an improvement of various deficits that closely correlate to symptoms in FXS patients. In contrast to the studies with antagonists, the reduction of mGlu5 receptor levels resembles a 50% receptor inhibition throughout the entire development of the FMR1 knock-out mice. The reduction of mGlu5 receptor levels by 50% in FMR1 knockout mice revealed a complete recovery of all phenotypes examined except macroorchidism [11], including: • Full rescue of deficient development of cortical plasticity in the FMR1 knockout (i.e., development of ocular dominance columns in the visual cortex) • Full rescue of deficient morphology of synaptic spines in the FMR1 knockout (i.e., increased total number and increased fraction of spines arrested in premature morphology) • Full correction of increased overall, basal rate of neuronal protein biosynthesis in the Fmr1 knockout • Full rescue of an exaggerated inhibitory avoidance extinction in the FMR1 knockout muchas gracias!!! => muchas gracias!!!
