épiderme du limbe formé de cellules à contours sinueux et de nombreux stomates de type anomocytique; épiderme des nervures comprenant des cellules parallélépipédiques à rectangulaires, allongées dans le sens de la nervure, de rares poils tecteurs (environ 500 µm de long) unicellulaires, raides, à parois épaissies et extrémité effilée, et de très rares poils sécréteurs, en massue, unicellulaires (environ 100 µm) entourés à la base d’une rosette de cellules annexes.

Abstract: Light and scanning electron microscopic studies and guard cell isolation techniques have confirmed the well-known ridges of the Equisetum stomatal apparatus as belonging to the subsidiary cells. Hitherto unknown features of the subsidiary cells such as the presence of a concentrated H2SO4-resistant region on the ridges and an interlocking mechanism for the closure of the aperture of the subsidiaries are described. These are presented as further evidence for the differences between the two subgenera of Equisetum. Boiling in dilute NaOH is shown to be a simple but effective means for the isolation of the guard cells in Equisetum as well as in several other plants with sunken stomata. Silicification of the outer layer of the epidermis makes cuticular isolation a difficult process by usual methods (treatments with enzymes or cellulose hydrolyzing reagents). Treatment with concentrated sulfuric acid followed by hydrofluoric acid results in the isolation of the cuticular membranes in Equisetum spp. and in similarly silicified grasses. Involvement of potassium ions in stomatal movements is indicated for two ferns and suggested for two species of Equisetum.

Résumé : Au moyen d'études en microscopie photonique et en microscopie électronique à balayage et de techniques d'isolement des cellules stomatiques, les auteurs ont confirmé que les crêtes de l'appareil stomatique d'Equisetum appartiennent aux cellules annexes. Les auteurs décrivent des caractéristiques nouvelles des cellules annexes tels que la présence d'une région résistante à H2SO4 concentré sur les crêtes et un mécanisme d'emboîtement pour la fermeture de l'ouverture des cellules annexes. Ces caractéristiques sont présentées comme différences additionnelles entre les deux sous-genres d'Equisetum. Une méthode simple mais efficace d'isoler les cellules stomatiques chez Equisetum et chez plusieurs autres plantes à stomates enfouis est de chauffer à ebullition dans NaOH dilué. La silicification de la couche extérieure de l'épiderme rend difficile l'isolement de la cuticule par les méthodes habituelles (traitements avec des enzymes ou des réactifs pouvant hydrolyser la cellulose). Un traitement à l'acide sulfurique concentré suivi par un traitement à l'acide fluorhydrique résulte en l'isolement des membranes cuticulaires chez Equisetum spp. et chez les graminées à épiderme silicifié.
