Wir bitten um Verständnis, dass eine FIX-Reservierung erst nach einer spesenfreien Anzahlung pro Zimmer erfolgt.

Spesenfreie Anzahlung doesn't make sense...free-of-charge deposit???

or are they trying to say you make a simple deposit and will not pay for the extras yet???

my second interpretation is: you make a deposit, which you will either receive back fully or in part if (and depending when) you cancel or it will be balanced off with your final hotel bill...

interest-free deposit?

Explanation:
Martin, this is exactly what you yourself suggested right from the beginning - which is why I was reluctant to post it as an answer.
I am not altogether happy with it though, because it doesn't really make it clear that the hotels want to avoid handling charges for themselves and not for their guests. Hence the addition in brackets

From what I gave gathered through googling, "spesenfrei" means that the payment method used for the deposit doesn't incur any charges for the hotel (e.g credit card charges). All the hotels I have checked on the internet provide bank details for payment incl. IBAN and BIC numbers so as to avoid these charges.
