Explanation:
It all depends on the context, but at a first glance, I took "天下の桜田門" as the object, not the subject. The subject should be we or I then.

Like, "Give me a break! No way that we could investigate the holy Metropolitan Police!".

I assume they are policemen at a much lower rank.

I don’t know the storyline, but I think your line “おいいい加減にしろ！天下の桜田門が捜査できるかってんだ” is said in irony. (Like, actually, the metropolitan police really can’t do a proper investigation because they’re either inept, or they can’t for other reasons.)
