II.2- Prélèvement des échantillons
Les tests à effectuer portent sur 3 catégories de produits :
1/ Produits finis lyophilisés
Le prélèvement effectué par CCCCC, constitué du nombre d’unités nécessaires à l’analyse est transmis tel quel à AAAA qui réalisera lui-même la dilution par un solvant approprié stérile et apyrogène avant l’injection.
2/ Produits finis liquides
Le prélèvement effectué par CCCCC, constitué du nombre d’unités nécessaires à l’analyse est transmis tel quel à AAAA qui réalisera lui-même la dilution éventuelle par un solvant approprié stérile et apyrogène avant l’injection.
3/ Matières premières sous forme de poudre
Le prélèvement effectué par CCCCC, constitué de la quantité nécessaire à l’analyse est transmis dans un récipient stérile et apyrogène à AAAA qui réalisera lui-même la dilution éventuelle par un solvant approprié stérile et apyrogène avant l’injection.
II.3- Modalités d’envoi, d’étiquetage et de réception des échantillons
Tous les échantillons à tester sont étiquetés de la manière suivante par CCCCC :

Les échantillons sont disposés dans des emballages garantissant l’intégrité des échantillons pendant le transport. La demande d’envoie est incluse dans le colis.
Sur la commande est indiqué le pays de destination du lot.
Le XXXXX est envoyé dans une boîte isotherme avec « freeze pack ».
Les échantillons sont envoyés à raison de 1 envoi ou plus par semaine à l’adresse suivante :

A réception du colis, AAAA envoie à CCCCC un fax d’accusé réception mentionnant éventuellement les prélèvements cassés nécessitant un remplacement ainsi que la planification d’envoi des résultats.
III- MISE EN ŒUVRE DES TESTS
III.1- Tests pyrogènes
III.1.1- Les animaux
Il s’agit de lapins blancs Néo-Zélandais originaires d’un établissement agréé, pesant entre 1.5 kg et 4.2 kg, stabulés 2 semaines au moins avant d’être utilisés.
III.1.2- Méthode utilisée
Les tests pyrogènes sont effectués selon les prescriptions générales de la Pharmacopée Européenne en vigueur et selon les spécifications particulières CCCCC. La Méthode d’analyse du AAAA figure en annexe 2.
Les spécifications CCCCC de chaque produit testé sont envoyées au AAAA à chaque mise à jour par CCCCC.
III.1.3- Compte rendu des résultats
Les calculs sont effectués à l’aide d’un logiciel pyrogène, les résultats sont édités sous forme d’un compte rendu informatisé (cf exemple annexe 3)
L’acceptation ou le refus du test sont faits selon la Pharmacopée Européenne sans aucune interprétation.
Les essais suivants un premier essai non satisfaisant sont refaits le plus tôt possible dans la limite des places disponibles, jusqu’à obtention d’un résultat interprétable selon la Pharmacopée Européenne.
Le résultat est transmis à CCCCC par Fax ou par e-mail et confirmé par le compte rendu envoyé par la poste.
III.1.4- délais
Les tests pyrogènes sont réalisés dans les 6 jours ouvrables maximum à réception des échantillons.
III.2- Toxicité anormale
III.2.1- Les animaux
Il s’agit de souris blanches mâles ou femelles pesant 17 à 22 g, origine DEPRE.
III.2.2- Méthode utilisée
Les tests de toxicité anormale sont effectués selon les prescriptions générales de la Pharmacopée Européenne en vigueur et selon les spécifications particulières CCCCC (Cf annexe 4).
Les spécifications CCCCC de chaque produit testé sont envoyées au AAAA à chaque mise à jour.
La Méthode d’analyse du AAAA figure en annexe 4.
III.2.3- Compte rendu des résultats
Le lendemain de la fin de l’essai le résultat de recherche de toxicité anormale est transmis à CCCCC par fax ou par e-mail puis confirmé par un compte rendu envoyé par la poste (cf exemple annexe 5)
III.2.4- délais
Les tests toxicité anormale sont réalisés dans les 8 jours ouvrables maximum à réception des échantillons.

II.2- Sampling
The tests to be carried out are on 3 product categories:
1/ Lyophilised finished products
The sample produced by CCCCC constitutes the number of units necessary for the analysis and are transmitted as such to AAAA who will carry out the required dilution with an appropriate sterile apyrogen solvent before injection.
2/ Liquid finished products
The sample produced by CCCCC, constitutes the number of units necessary for the analysis and is transmitted as such to AAAA who will carry out the required dilution with an appropriate sterile apyrogen solvent before injection.
3/ Raw materials in powder form
The sample produced by CCCCC, constitutes the number of units necessary for the analysis and is transmitted in a sterile and apyrogen container to AAAA who will carry out the eventual dilution with an appropriate sterile apyrogen solvent before injection.
II.3- Means for the despatch, labelling and reception of samples
All samples to be tested are labelled in the following manner by CCCCC:

The samples are arranged in a container guaranteeing the integrity of the samples during transport. Test details are included in the parcel.
The country of destination for the batch is indicated on the order.
The SXXXXX is sent in an isotherm box with ice packs.
The samples are to be sent, 1 or more each week to the following address:

On reception of the parcel, AAAA sends a fax to CCCCC confirming reception noting any breakages that require replacing as well as the timetable for returning the results.
III- TEST PROCEDURE
III.1- Pyrogen tests
III.1.1- The animals
These are New Zealand white rabbits, from an approved establishment, weighing between 1.5 and 4.2 kg that have been kept on site at least two weeks prior to being used.
III.1.2- Methods used
The pyrogen tests are carried out in accordance with the general Eu.Ph. rules in force and to specific CCCCC specifications. The AAAA analytical method is shown in appendix 2.
The CCCCC specifications for each product tested will be sent to AAAA every time it is updated by CCCCC.
III.1.3- Results report
The calculations are made with the help of a pyrogen program. The results are to be produced in a computerised printout (see example in appendix 3).
Acceptance or refusal of the test is made according to the Eu.Ph. without any interpretation.
Trials carried out after initial unsatisfactory trials must be re-done as soon as possible, within the limits of availability, until interpretable results have been obtained according to the Eu.Ph.
The results are sent to CCCCC by fax or by e-mail and confirmed with the written report sent by post.
III.1.4- Delay
The pyrogen tests are carried out within a maximum of 6 days after reception of the samples.
III.2- Abnormal toxicity
III.2.1- The animals
These are white mice, either male or female, weighing between 17 and 22g, from the DEPRE.
III.2.2- Methods used
The abnormal toxicity tests are carried out in accordance with the general Eu.Ph. rules in force and to specific CCCCC specifications. (see appendix 4).
The CCCCC specifications for each product tested will be sent to AAAA every time it is updated by CCCCC.
The AAAA analytical method is shown in appendix 4.
III.2.3- Results report
The morning after the end of the test of abnormal toxicity, the results are sent to CCCCC by fax or by e-mail and confirmed with the written report sent by post (see example in appendix 5).
III.2.4- Delay
The abnormal toxicity tests are carried out within a maximum of 8 days after reception of the samples.
