Il 28 Settembre 1893 Giovanni Segantini scriveva da Maloja a Vittore Grubicy, suo mercante d’arte a Milano: «Ciò che rapisce e affascina il mio spirito, è l’amore illimitato che provo per la natura. [...] Quando le circostanze mi costringono ad ignorare la natura per guardare dentro di me, come nel caso del quadro L’angelo della vita, al quale sto appunto lavorando, succede che mi sento continuamente ed ininterrottamente tormentato da rimorsi di coscienza, che mi lacerano come se stessi compiendo una cattiva azione, anche se so di fare la cosa giusta » Nel concepire contenuti simbolisti, l’artista non solo doveva affrontare le difficoltà dell’immaginazione figurativa, ma anche vincere una «tensione di principio verso la dedizione alla realità visibile» . Tramite la meticolosa osservazione della natura, Segantini puntava a rappresentarne fedelmente le leggi eterne e le profonde verità, alle quali tutte le creature – uomini ed animali – sono soggette. In questa sezione è presentato il programma di breeding svoltosi negli anni 2001-2005 al fine di creare nuovi portainnesti che portino il carattere di resistenza associato agli altri tratti di interesse commerciale. I donatori di resistenza Malus sieboldii, le selezioni apomittiche di prima generazione (4551, 4608) e le selezioni apomittiche di seconda generazione (C1907, D2118, D2212, H0801, H0909, Gi477/4) si rivelano troppo vigorosi per essere utilizzati come portainnesti di uso commerciale. L’analisi con i marcatori SSR ha inoltre permesso di stabilire delle ipotesi sul livello di ploidia dei parentali e delle progenie. Tali ipotesi sono state confermate da uno studio citofluorimetrico. La produzione di frutti e di semi in alcune combinazioni di incrocio può essere speigata in parte da questi peculiarità legate alle caratteristiche di apomissia e poliploidia portate del genitore resistente. I primi risultati delle osservazioni fenotipiche, ottenuti dopo 1-2 anni dall’infezione dei semenzali, hanno dimostrato come il carattere della resistenza sia stato ereditato dalle progenie. Le popolazioni segreganti identificate in questo lavoro sono attualmente impiegate allo scopo di studiare le basi genetiche della resistenza ad Apple proliferation e per la costruzione di mappe di linkage utilizzate per identificare geni associati alla resistenza. SECTION B: BREEDING PLAN
This section describes the breeding plan carried out in 2001-2005 to create new grafts that would carry the resistance trait (along with other traits of commercial interest). Malus Sieboldii resistance donors, apomitic selections of the first generation (4551, 4608) and apomitic selections of the second generation (C1907, D2118, D2212, H0801, H0909, Gi477/4) all proved to be too vigorous to be used as grafts for commercial purposes. The over 3000 seedlings obtained were typed using codominant microsatellite (SSR) markers in order to establish their true pedigree and to characterize new genetic resources for improving the apple tree genetically. Analyses using a reduced number of SSRs, which were suitably selected for their polymorphism, identified different classes of seedlings that were present in different proportions in the various crosses: seedlings that were genetically identical to the mother; seedlings deriving from probable self-pollination; and two classes of individuals with the alleles of the male parent at all loci. Out of the cross combinations that used an apomitic plant as the seed bearer, 67% of the seedlings exhibited a genotype that was the same as the mother’s and thus were not useful for breeding. Analysis using SSR markers also allowed us to postulate several hypotheses regarding the level of ploidy of the parent specimens and their progeny. These hypotheses were confirmed by cytofluorimetric study. The production of fruit and seeds in certain cross combinations can be explained in part by these unique features, which are associated with the traits of apomixia and poliploidia carried by the resistant parent. The initial results of phenotypic observations made 1-2 years after seedling infection have shown that the resistance trait was inherited by the progeny. The segregated populations identified in this study are currently being used to examine the genetic basis for resistance to apple proliferation and to create maps of linkages for identifying the genes associated with resistance. Additional studies on resistant genotypes will probably be carried out over the next few years in order to evaluate their operative agronomic value.
