Apoptosis, a form of cell death, is a fundamental process for the development and maintenance of multicellular organisms that promotes the removal of damaged, senescent or unwanted cells. Induction of cancer cell apoptosis is an important strategy of anticancer therapy. In this study, we examined if melatonin, the main secretory product of the pineal gland, inhibited the growth of prostate cancer cells (LNCaP) and promoted apoptosis via mitogen-activated protein kinases (MAPKs), which are closely associated with apoptosis and survival. Melatonin treatment significantly inhibited the growth of LNCaP cells in a dose- and time-dependent manner. It clearly induced both an early stage of apoptosis (propidium iodide(-), FITC Annexin-V( )) and a late apoptosis/secondary necrosis (propidium iodide( ) and FITC Annexin-V( )), which indicated induction of serial stages of apoptosis in cells. Moreover, melatonin markedly activated c-JUN N-terminal kinase (JNK) and p38 kinase, whereas extracellular signal-regulated kinase (ERK) was not responsive to melatonin. Treatment with MAPK inhibitors, PD98059 (ERK inhibitor), SP600125 (JNK inhibitor) and SB202190 (p38 inhibitor), confirmed that melatonin-induced apoptosis was JNK- and p38-dependent, but ERK-independent.

В данном исследовании мы проверяли, препятствует ли мелатонин, основной продукт эпифиза мозга, росту раковых клеток предстательной железы линии LNCaP и способствует ли он апоптозу, опосредованному митоген-активируемыми протеинкиназами (MAPKs), которые тесно связаны с апоптозом и выживанием. Обработка мелатонином существенно подавляла рост клеток линии LNCaP, также была показана зависимость эффекта от дозы и времени. Совершенно очевидно, что она индуцировала раннюю стадию апоптоза (пропидиум йодид (-), FITC аннексин-V( )) и поздний апоптоз/вторичный некроз (пропидиум йодид ( ), FITC аннексин-V( )), что указывает на индукцию различных механизмов апоптоза в клетках. Procedure and system suitability test:
The column is equilibrated by injection of solvent. The reference solution (a) is injected three times.
The standard deviation for the peak areas of the analyzed compound base on replicate injection of reference solution (a) is not allowed to exceed n%.
The peak-to-valley ratio between the analyzed compound main peak and the impurity A in the chromatogram obtained with reference solution (b) has to be at least x.
In case that the requirements of the system suitability test are not met, then the time for conditioning of the system should be prolonged. If this measure is not efficient, then further adjustments of the chromatographic conditions within the provisions of chapter 2.2.46 (Ph. Eur.) can be performed. If necessary, the separating column is replaced or the complete chromatographic system must be checked.
Evaluation:
The test is evaluated according to the method of the external standard. The mean value of the peak area calculated from three injections as well as chromatograms of reference solution (a) are used.
All impurities are evaluated against the peak area of the analyzed compound peak in the chromatogram obtained with reference solution (a).

Оценка результатов анализа:
Данный анализ выполняется по методy внешнего стандарта. Для этого используют среднее значение площади пика, рассчитанное после трех введений, а также хроматограммы контрольного раствора (a).
Все примеси оцениваются по сравнению с площадью пика анализируемого вещества на хроматограмме контрольного раствора (a).

Vitamin E is a term that describes a group of compounds with similar yet unique chemical structures and biological activities. One interesting property possessed by certain vitamin E compounds – namely, δ-tocotrienol, RRR-α-tocopheryl succinate [vitamin E succinate (VES), a hydrolysable ester-linked succinic acid analogue of RRR-α-tocopherol], and a novel vitamin E analogue referred to as α-TEA (α-tocopherol ether linked acetic acid analogue, which is a stable nonhydrolyzable analogue of RRR-α-tokopherol) - is their ability to induce cancer-cell by not normal cells to undergo a form of cell death called apoptosis. In contract, the parent compound, RRR-α-tocopherol, also referred to as natural or authentic vitamin E and known for its antioxidant properties, does not induce cancer-cell apoptosis. Efforts to understand how select vitamin E forms can induce cancer cell to undergo apoptosis have identified several nonantioxidant biological functions, including restoration of pro-death transforming growth factor-β and Fas signaling pathways.

Если бы врачи, борющиеся за здоровье и жизнь своих пациентов, отказывались от лечения тех, кто сам внес основной вклад в свой недуг, большую часть больниц можно было бы закрыть за ненадобностью. И дело не только в невнимании к себе, заканчивающемся визитом к стоматологу только в случае сильной зубной боли, или в чрезмерной любви к острым ощущениям, заканчивающейся койкой в травматологическом.
Один из самых серьезных аргументов в пользу здорового образа жизни – сахарный диабет, 90% случаев которого приходятся на диабет 2 типа, развивающийся с возрастом из-за переедания, ожирения и наследственных факторов. Помимо лекарственной терапии в таком случае отлично помогает низкокалорийная диета с минимумом жиров и углеводов.

Однако попытки заменить макароны с салом постной говядиной в бобах тоже небезопасны.

Problem here is not only in the negligence to one's health, which leads to dental visits only in the case of an acute toothaches, or in the thrill seeking, which often concludes in a Trauma Center visit.
One of the gravest arguments in favor of a healthy lifestyle is diabetes mellitus, with type II diabetes accounting for 90% of all cases, which develops with age due to overeating, obesity, and hereditary factors. Apart from the drug therapy, a low-calorie diet with the minimal amount of fat and carbohydrates will help greatly in that case.

However, attempts to substitute spaghetti and bacon with lean beef and beans are not safe, either. As it turns out, proteins are also able to contribute to the insulin sensitivity loss in the cells.

Cristopher Newgard of the Sarah W. Stedman Nutrition & Metabolism Center and his colleagues has noticed that the diet of obese people - even those trying to control their calorie intake - is rich in diverged polypeptides. It is this fact that led scientists to the conclusion, that proteins contribute their fair share to the development of diabetes. The authors of the Cell Metabolism publication were able to prove their hypothesis using laboratory mice as test subjects; however, they still refrain from giving advice regarding the remedial diets for patients.

Insulin is a key molecule governing metabolic functions throughout human organism.
After the food intake and glucose is being transferred from the intestinal wall into the blood, the pancreas ejects insulin into the blood stream, thus stimulating all our cells to increase their energy consumption. The breakdown of fats and proteins slows down, while the liver and muscle tissue begin to synthesize glycogen from the excess sugar. To counter this system, other signal molecules, including adrenaline and stress hormones, suppress insulin production.
