The substrate stock solution consisted of 20 mM each of phenacetin, diclofenac, diazepam, bufuralol, midazolam and 7-hydroxycoumarin in 1% dimethyl sulphoxide. Initially 7-ethoxycoumarin was used instead of 7-hydroxycoumarin in incubations with rat, human and dog hepatocytes. The stock solution was stored in aliquots at -20@C until incubation. All incubations were performed as duplicates in 96-well deep microplates (Treff Ag, Degersheim, Switzerland). The hepatocytes were diluted in Krebs-Henseleit buffer and 90 ml of the cell suspension were added to 10 ml of the cocktail stock solution so that the final hepatocyte concentration was 106 cells ml-1, and the concentration of each enzyme substrate was 2 mM. Incubations were performed at 37C under an atmosphere of 5% CO2/95% air and the plates were shaken gently.
The human, dog and minipig hepatocytes were incubated with the cocktail for 0, 15, 30, 45, 60 and 90 min, while the rat hepatocytes were incubated with the cocktail for 0, 5, 15, 30, 45 and 60 min. The reactions were stopped by the addition of 100 ml ice-cold acetonitrile. The plates were centrifuged (1900 g, 5 min) and aliquots of the supernatant analysed by LC-MS/MS.
The cocktail compounds were screened for possible inter-substrate interactions in human hepatocytes by also incubating each compound individually. The results thus obtained were compared with the corresponding results obtained from incubations with the cocktail.
Inhibition with ketoconazole
Human hepatocytes were incubated with the cocktail, as described above, together with 1 mM ketoconazole. The experiment was performed four times with hepatocytes from one or two individuals in each experiment, with a total number of five individuals (Lots CYE, ENR, HRK, OCF and OQD). All samples were treated in exactly the same manner as described for the hepatocytes incubated without the inhibitor.

A specification of the format of the container (the higher-level elements that provide information about the file as a whole and about entries). In TMX, an entry consisting of aligned segments of text in two or more languages is called a Translation Unit (the element).
A specification of a low-level meta-markup format for the content of a segment of translation-memory text. In TMX, an individual segment of translation-memory text in a particular language is denoted by a element. See the section Content Markup for more details.
TMX offers two levels of implementation:
Level 1 (Plain Text Only) - Support for the container only. The data inside each element is plain text, without Content Markup. This level is enough when the data do not have inline codes, for example software messages. It is not sufficient for documentation-type formats.
Level 2 (Content Markup) - Support for both container and content. The application uses the TMX Content Markup, to allow any other tools supporting also TMX Level 2 to re-create the translated version of an original document by using only the TMX document.
The information on how the translation units have been segmented is carried in a separate document: a Segmentation Rules eXchange (SRX) file.

1.1. XML Compliance
TMX is XML-compliant. It also uses various ISO standards for date/time, language codes, and country codes. See the References section for more details.
TMX files are intended to be created automatically by export routines and processed automatically by import routines. TMX files are "well-formed" XML documents that can be processed without explicit reference to the TMX DTD. However, a "valid" TMX file must conform to the TMX DTD, and any suspicious TMX file should be verified against the TMX DTD using a validating XML parser.
