Protocol Title
A Phase III Open Label, Randomized Two-Parallel-Arm Multicenter Study of E7389 versus Capecitabine in Patients with Locally Advanced or Metastatic Breast Cancer Previously Treated with Anthracyclines and Taxanes and Refractory to the Most Recent Chemotherapy
For the duration of response, Kaplan-Meier plots 95% will be provided along with the median and the confidence interval. Tumor response data utilized for primary analysis of PFS, ORR and duration of response will be obtained from an independent review of the imaging scans. In addition, these analyses will be performed using the investigators’ determination of response. Response will be assessed according to RECIST criteria.
Baseline tumor assessments, consisting of CT or MRI scans of the chest, abdomen and pelvis and any other areas of suspected disease, as well as photographs of skin lesions being measured as target lesions should be performed within 28 days prior to starting study treatment. Areas where disease was found at Baseline will then be scanned/photographed every second cycle (starting Cycle 2) between Days 15 and 21, or sooner if there is evidence of progressive disease. Any new areas of suspected disease identified at these time-points should also be scanned/photographed.

Per la durata della risposta, verranno forniti tracciati di Kaplan-Meier al 95% insieme alla mediana ed all'intervallo di confidenza. I dati sulla risposta tumorale utilizzati per l’analisi primaria di sopravvivenza libera da progressione, tasso di risposta obiettiva e durata della risposta verranno ottenuti da una revisione indipendente della diagnostica per immagini. Inoltre, queste analisi verranno eseguite utilizzando la determinazione della risposta dei ricercatori. La risposta verrà valutata in base ai criteri RECIST.
Entro 28 giorni prima dell’inizio del trattamento in studio dovranno essere eseguite valutazioni del tumore al basale, che consisteranno in TC o RMN del torace, dell’addome e della pelvi e qualsiasi altra area di sospetta malattia, come pure in fotografie delle lesioni cutanee misurate come lesioni bersaglio. Le regioni in cui verrà riscontrata la malattia al basale verranno sottoposte a scansione/fotografate ogni due cicli (a cominciare dal Ciclo 2) tra i Giorni 15 e 21 o prima, in caso di evidenza di malattia progressiva. Die Listeriose ist eine bei Mensch und Tier auftretende gefährliche Infektionskrankheit, die in rund 30 % der Fälle einen tödlichen Verlauf nimmt. Verursacher ist vor allem das Bakterium Listeria monocytogenes, das in der Umwelt weit verbreitet ist und unter Umständen unsere Lebensmittel kontaminiert.
Nicht jeder, der die Keime aufnimmt, muss jedoch erkranken. Gesunde Menschen erkranken in der Regel nicht an Listeriose.
Als Risikogruppen gelten Schwangere und ihre un- oder neugeborenen Kinder sowie immungeschwächte Personen wie Krebs- und AIDS-Kranke, Alkoholiker, und häufig auch alte Menschen.
Typische Krankheitsbilder einer Listeria-Infektion sind Blutvergiftung (Sepsis), Hirnhautentzündung (Meningitis) und grippeähnliche Symptome wie Fieber, Muskelschmerzen und Erbrechen. Vereinzelt können auch Infektionen wie Gelbsucht (Hepatitis) und Hauterkrankungen auftreten.

La listeriosi è una pericolosa malattia infettiva che compare nell’uomo e nell’animale e che ha un decorso letale ca. nel 30% dei casi. La causa è soprattutto il batterio Listeria monocytogenes che è ampiamente diffuso nell’ambiente e contamina i nostri alimenti in determinate condizioni. Non tutti coloro che ingeriscono questi germi si ammalano, però. Gli individui sani generalmente non si ammalano di listeriosi.
Sono a rischio le donne in gravidanza e il proprio feto o i neonati, come pure le persone immunocompromesse come malati di cancro e AIDS, alcolisti e spesso anche gli anziani.Il quadro clinico tipico di un’infezione da listerie è caratterizzato da avvelenamento del sangue (setticemia), infiammazione delle meningi (meningite) e sintomi similinfluenzali come febbre, dolore muscolare e vomito. In casi singoli, possono manifestarsi anche infezioni come itterizia (epatite) e affezioni della cute.
