Source text - Polish Wykrywanie peroksydazy 1. Podsuszony preparat utrwalamy w roztworze formaldehyde-etanolowym (1 część formaliny + 9 części 96° alkoholu etylowego) przez 30 min. 2. Preparat spłukujemy wodą bierzącą. 3. Preparat umieszczamy na 5 min w roztworze benzydyny (szcyptę tego związku rozpuszczamy w 12 ml 96° alkoholu, a następnie dopełniamy wodą destylowaną do 20 ml). Dodajemy 0,04 ml świeżej wody utlenionej. 4. Preparat spłukujemu ponownie wodą destylowaną i podbarwiamy barwnikiem Giemsy przez 15 min. Wyniki. Dojrzałe granulocyty obojętnochłonne barwią się na brązowo. Poza opisanymi wyżej metodami używa się niekiedy do barwienia preparatów komórkowych róźnych odmian techniki hematoksylina- eozyna, stosowanych rutynowo w pracowniach histologicznych. Dobre obrazy można uzyskać przy barwieniu hematoksyliną Meyera. Niektórzy zalecają hematoksylinę żelizistą Heidenhaina, która daje dobre obrazy struktur wewnątrzkomórkowych i wyrazisty rysunek chromatyny jądrowej. To ostatnie barwienie winno byc prowadzone pod kontrolą w mikroskopie.	Translation - English Peroxidase detection 1. Fix the dehydrated preparation in a solution of formaldehyde-ethanol (1 part formaline: 9 parts 96% ethyl alcohol) for 30 minutes. 2. Rinse in running tap water. 3. Place in a solution of benzidine for 5 minutes (dissolve a pinch of the compound in 12 mL of 96° alcohol, then fill to 20 mL with distilled water). Add 0.04 mL of fresh hydrogen peroxide. 4. Rinse again with distilled water and stain with Giems stain for 15 minutes. Results: Mature neutrophilic granulocyte stain brown. For cellular preparation staining different hematoxylin-eosin techniques can be used other than the ones just described. These are routinely carried out in histological laboratories. Staining with Mayer’s hematoxylin can produce good images. Some recommend Heidenhain’s iron hematoxylin which yields good intracellular structure images and a clear picture of the nuclear chromatin. This last staining should be performed under microscopical control.
