Source text - English Generally, the colored derivative should have a high extinction coefficient, ε = 104, to give good results. Ideally, the substrate organic compound for the colored derivative should be of low toxicity and be a crystalline solid with a high degree of purity. If an acid, such a substrate could function as a primary standard for an aqueous or alcoholic base titrant, so that a known weight of it can be analyzed by standard alkalimetric techniques. The colored indicator method for these primary standards should then be applied to some freshly prepared organometallic compound such as n-butyllithium which has been assayed by an independent method, such as the modified Gilman method and has been thus shown to contain less than 1% of free base (> 99% carbon-bound lithium). The values obtained by the two methods should not vary by more than ±1% from each other and, of course, reproducibility should also be well within this bias value.

Source text - English The membrane covered oxygen sensor (Clark principle) contains a Pt cathode and an Ag/AgCl anode. Both electrodes and
the electrolyte are separated by the membrane from the measuring medium. The membrane is only permeable to molecular
oxygen. Therefore disturbing components contained in the measuring medium cannot influence the function of the electro-
chemical measuring cell. Because of the external polarisation voltage of 700 mV across Pt cathode and Ag/AgCl anode the
oxygen diffusing through the membrane is reduced to hydroxyl ions. At the anode Ag+ions are generated (because of oxidation) reacting with the Cl-ions of the electrolyte solution to AgCl. The generated diffusion current is a dimension for the oxygen concentration.
The oxygen sensor consists of shaft, basic body and membrane head (figure 2). The shaft of the special versions MF 41 and
MF 43 is equipped with hex nut PG 13.5 for installing the probe in the process housing. In addition to the probes with fixed
cable, special versions with a multi pin plug head and separated cable connections are available. Furthermore the oxygen
sensor contains a Pt cathode, an Ag/AgCl anode and one ore two temperature probes. The oxygen permeable polymeric
membrane is integrated in the membrane head. The rugged multi-layer membrane allows reliable measurements for a long
period without changing membrane head and electrolyte.

Source text - English ХХХ is a specific vaccine against blue pus bacillus Pseudomonas aeruginosa.
ХХХ is recommended for use when there is a risk of P. aeruginosa infection and sepsis, particularly in patients with burns. ХХХ is used prophylactically to induce active immunity to P. aeruginosa infections. Therapeutically it is used when P. aeruginosa infection had already been contracted, in order to enhance specific immunity and lower the risk of bacteraemia and sepsis.
P. aeruginosa infections are usually life-threatening conditions in patients with dysfunctional immune system occurring in neoplasm disease, immunosuppressant treatment, burns and other devastating diseases. Since the efficacy of P. aeruginosa treatment with antibiotics is low, attention was paid to immunotherapy, and particularly to a possibility of active and passive immunization of patients at risk of P. aeruginosa infection or already suffering from this disease.
A vaccine for active immunisation had been under development since the mid-sixties and research focused on different methods of its preparation.
Drug substances of ХХХ vaccine are structural and extracellular antigens present in supernatant of heat-inactivated Pseudomonas aeruginosa strains representing immunotypes 1-7, according to Fisher, Devlin, Gnabasik classification. The strains are cultured on synthetic Davis medium.
No excipients are added during drug product manufacture. A preservative, phenol, present in the drug product is added during manufacture of drug substances.
In order to enhance immunogenicity of ХХХ vaccine and prolong exposure of organisms to be immunised to the antigen, an attempt was made to adsorb vaccine to aluminium hydroxide. Adsorption was hindered by bacterial mucus present in supernatants. A series of experiments was performed to optimize adsorption and to develop a procedure of mucus removal from the culture. The most efficient method of mucus removal from the culture was lowering of pH of P. aeruginosa culture to pH 5 and then, heating of culture at 100oC for 30 min. This procedure allowed to obtain a non-toxic, immunogenic and properly aluminium-hydroxide adsorbable vaccine.
In order to lower the toxicity of experimental vaccines, it was decided to inactivate strains constituting the vaccine by 0.3% formalin.
Vaccine prepared from formalin-inactivated supernatants of P. aeruginosa strains was immunogenic but toxicity was not lowered to the expected level. Due to high formaldehyde content in the vaccine and inability to carry out aluminium hydroxide adsorption, studies of this vaccine were discontinued.

XXX рекомендована к применению пациентам с риском заражения P. aeruginosa и возникновения сепсиса, в частности ожоговым пациентам.
XXX используют также для профилактики и усиления активного иммунитета против P. aeruginosa.
Данную вакцину применяют после контакта с инфекцией P. aeruginosa для усиления специфического иммунитета и снижения риска бактериемии и сепсиса.
Заражение P. aeruginosa опасно для жизни пациентов с дисфункцией иммунной системы при опухолевых заболеваниях, принимающих иммунодепрессанты, для ожоговых и других тяжелобольных пациентов. Лечение P. aeruginosa антибиотиками не эффективно, поэтому большое внимание уделяют иммунотерапии и, в частности, активной и пассивной иммунизации пациентов с риском развития синегнойной палочки или уже заболевших.
Вакцину для активной иммунизации разрабатывают с середины 60х годов, исследования сфокусированы на различных методах подготовки вакцины.
Активное вещество вакцины ХХХ – это структурные и внеклеточные антигены, присутствующие в супернатанте термоинактивированных штаммов Pseudomonas aeruginosa с 1-7 иммунотипами в соответствии с классификацией Фишера-Девлина-Гнабасика.
Штаммы культивируют на синтетической питательной среде Дэвиса.
При производстве лекарственного препарата не добавляют эксципиенты.
В качестве консерванта в лекарственном препарате присутствует фенол, добавляемый при производстве активного вещества.
В целях повышения иммуногенности вакцины ХХХ и увеличения срока защитного действия были попытки адсорбировать вакцину на гидроксид алюминия. Адсорбции препятствовали бактериальные слизи в супернатантах. Провели серию экспериментов по оптимизации адсорбции и разработке методов удаления слизи из культуры. Бактериальную слизь можно эффективно удалить при снижении рН культуры P. aeruginosa до 5 и дальнейшим нагревание культуры при 100оС в течение 30 минут. Благодаря этому методу образуется нетоксичная, иммуногенная и хорошо адсорбируемая на гидроксиде алюминия вакцина.
В целях снижения токсичности экспериментальных вакцин, было предложено инактивировать штаммы 0.3% формалином. Вакцина, полученная из инактивированных формалином штаммов – иммуногенна, но ее токсичность не ниже расчетного уровня. Разработку данной вакцины прекратили, так как она содержала высокие количества формальдегида и плохо адсорбировалась на гидроксиде алюминия.

I graduated from the Chemical Dept of Belorussian State University in 2013.
