Introduction
Angiogenesis is the process of new blood vessels formation from existing vasculature through budding and the growth of tubes formed initially of endothelial cells. This process is controlled by a complex interaction of factors that either stimulate or inhibit angiogenesis.
One of the most important of these factors is vascular endothelial growth factor (VEGF), which is a pro-angiogenic factor. VEGF has multiple roles in controlling the proliferation and survival of endothelial cells. Importantly, VEGF is a critical factor in angiogenesis during tumour growth. Tumour vascularisation is an essencial part of tumour progression and so a target for therapy.
To understand why avastin (bevacizumad) was developed and how it exerts its effects, you must be familiar with the process of angiogenesis in health and in cancer and the role of VEGF within that process.

Introdução
A angiogénese consiste no processo de formação de vasos sanguíneos novos a partir de outros pré-existentes, através da proliferação e crescimento de capilares formados a partir de células endoteliais. Este processo é controlado por uma complexa interacção de factores estimuladores e inibidores da angiogénese.
Destes factores, um dos mais importantes é o Factor de Crescimento do Endotélio Vascular (VEGF) que é um factor de efeito pró-angiogénico. Tem múltiplas funções no controlo da proliferação e sobrevivência das células endoteliais. É de sublinhar que o VEGF é um factor crítico na angiogénese durante o crescimento de tumores. A vascularização dos tumores é uma etapa essencial da progressão tumoral e como tal constitui um alvo terapêutico. Para se compreender por que se desenvolveu o avastin (bevacizumab) e a forma como este actua, torna-se necessário conhecer como se processa a angiogénese em tecidos normais e cancerosos, bem como a função intrínseca do VEGF neste processo.
