PharmaMar has presented Phase I and Pharmacokinetic results of Kahalalide F at the 40th Annual Meeting of the American Society of Clinical Oncology (ASCO), taking place in New Orleans from 5-8 June.
A poster presented by Dr. Jos H. Beijnen et al, The Netherlands Cancer Institute, detailed the results from: "A Phase I and Pharmacokinetic study of Kahalalide F given intravenously over one hour, during five consecutive days every three weeks in patients with advanced or metastatic androgen refractory prostate cancer."
The objectives of the study were to determine the safety and maximum tolerated dose, explore the pharmacokinetics and document the anti-tumour activity of Kahalalide F. The setting was patients suffering from androgen independent prostate tumours. Thirty-two patients were registered with a median age of 68 years. They had metastases to the bone, lymph nodes, liver and/or lungs and had received prior hormone therapy, radiotherapy and/or surgery.
Overall, hematologic toxicities were negligible and were not clearly dose-related. Using the NCIC-CTC (National Cancer Institute of Canada - Common Toxicity Criteria) classification, G1 anemia, thrombocytopenia and neutropenia were observed in 22 patients. G2 anemia and neutropenia occurred in 8 patients. There were no G3 or G4 hematologic toxicities. Other side effects observed were nausea & vomiting, fatigue, hematuria and hypersensitivity reactions, none of which were at G4. The major drug related toxicity was rapidly reversible transaminase elevation at G3/G4.
The conclusion of the study was that Kahalalide F can be administered safely as a 1-hour intravenous infusion during five consecutive days at a dose of 560 mg/msquared/day once every three weeks. Future studies in hormone refractory advanced prostate cancer are warranted.
Dr Miguel Angel Izquierdo, Director of Clinical Development at PharmaMar, said:
"These results are consistent with previous studies undertaken with Kahalalide F. We are impressed by the safety profile of the product and are on track to continue the development of this compound."

Het onderzoek werd gedaan onder patiënten met hormoonongevoelig prostaatkanker.Tweeëndertig patiënten met een mediane leeftijd van 68 jaar namen aan het onderzoek deel. De patiënten hadden uitzaaiingen naar het bot, de lymfeklieren, de lever en/of de longen, en waren reeds behandeld met hormoontherapie, bestraling en/of geopereerd.
Het onderzoek toonde aan dat hematologische toxiciteit verwaarloosd kon worden, en niet duidelijk met de dosis samenhing.Gebruikmakend van het classificatiesysteem van het Nationale Kankerinstituut in Canada (NCIC-CTC - National Cancer Institute of Canada - Common Toxicity Criteria), werd G1 anemie, trombocytopenie en neutropenie bij 22 patiënten vastgesteld. G2 anemie en neutropenie kwam voor bij 8 patiënten.
Er werd geen G3 of G4 hematologische toxiciteit gevonden. Andere bijwerkingen, zoals misselijkheid en braken, vermoeidheid, hematurie en overgevoeligheidsreacties bereikten geen G4 niveau. De belangrijkste druggerelateerde toxiciteit was een snel-reversibele transaminase op het niveau van G3/G4.
De conclusie van de studie was dat Kahalalide F veilig in de vorm van een 1-uur durend infuus van 560 mg/m²/dag gedurende 5 achtereenvolgende dagen kan worden toegediend, en iedere drie weken kan worden herhaald. Toekomstig onderzoek naar hormoonrefractair gevorderd prostaatkanker is aanbevolen.

I have a drive to be exact and look for a good understanding of the context of information to get the right message across. With an interest in a wide range of topics, an eagerness to familiarize myself with new information, and always applying a “scoping” attitude, I have an expanded view of what translation is about. My approach to translation:
Translation comes from Trans Relatus, meaning “Beyond Bringing Back.” Translation needs to go beyond correctly returning original words in another language. It may include translating into a different cultural context with different cultural facts, as with “localization” of websites. But that holds true for more traditional texts as well. Translation includes “interpretation” – and finding the right words for what the author intends to say. In other instances it may be essential to keep flavors of local elements, and skillfully bring the reader over into it, by being knowledgeable and sensitive about associated facts and history of words. Translation usually requires good knowledge of the translated topic and most definitely goes “beyond bringing back.“
Background in Psychology/Business/Management/Systems/Social Sciences/Philosophy/Spirituality:
I studied psychology at the University of Tilburg and received my BA in 1983. Highest graded courses were Psychology of Religion, Historical Psychology and Philosophical Anthropology. I did master’s courses in Clinical and Theoretical Psychology at the University of Utrecht, then went back to Tilburg to study Organizational Psychology, focusing on Organizational Learning (as requirement in a continuous changing environment) and Soft Systems Methodology (as a systems approach to the learning and problem solving process). I wrote a teaching manual on SSM for O&E, Philips, Eindhoven, and other study paper topics were on critical psychology, anti-psychiatry, science, sexual abuse in psychotherapy, Freud’s revision of his early seduction theory, on narcissism according to Alice Miller and Christopher Lasch. I enjoy teaching, explaining complex concepts, and was awarded a student teaching assistantship at the University of Utrecht in “Philosophy of Science.”
