Genotoxicity
The mutagenic potential of a range of nicotine concentrations has been studied using a variety of in vitro and in vivo genotoxicity tests. The results of these studies indicate that nicotine was not mutagenic in the appropriate assays. However, some evidence of chromosomal damage has been observed in other tests. For an in depth review of previously reported studies on the mutagenic potential of nicotine and its metabolites, the reviewer is referred to Part IIID of the Toxico-Pharmacological Documentation Summary of the NiQuitin CQ transdermal patch application.
Three additional studies of nicotine’s potential mutagenic and genotoxic effects have been published in the scientific literature since the submission of the NiQuitin CQ transdermal patch application. One of these studies evaluated the potential for nicotine to damage the DNA of human lymphocytes and lymphatic cells of human palatine tonsils using the alkaline single cell microgel electrophoresis (Comet) assay. In this assay, lymphocytes from human whole blood and tonsillar cells were isolated and prepared as cell suspensions. The cell suspensions were then incubated with nicotine at concentrations ranging from 0.125 to 4 mM for 60 minutes. N-methyl-N’-nitro-N-nitrosoguanidine (MNNG), at a concentration of 0.02 mM, was used at the positive control and phosphate buffered saline, at concentrations from 1 to 10%, served at the negative control. The measure for DNA damage used in this assay was the degree of DNA migration, expressed as the Olive tail moment, which represents the percentage of DNA in the tail and the tail length. Olive tail moment is a measure of possible DNA single strand breaks, alkali labile sites and incomplete excision repair sites. The results of this assay revealed that exposure to nicotine for 60 minutes at 0.125, 0.25, 0.5, 1, 2, and 4 mM induced a statistically significant dose-dependent increase of DNA migration up to 3.8-fold and 3.2-fold in tonsillar cells and lymphocytes, respectively. The lowest concentration eliciting a significant effect was 0.5 mM nicotine. This effect was confirmed in a second series of experiments using nicotine of high purity from two different suppliers. There were no significant differences between both series excluding artifacts due to the source of nicotine. The investigators of this assay concluded that the data indicate that nicotine causes a significant direct genotoxic effect in human target cells in vitro.13
Another recently published study examined the effect of nicotine on DNA damage induction and apoptosis in human gingival fibroblasts using an in vitro cytokinesis-block micronucleus test. The results of the study indicated that treatment of human gingival fibroblasts with nicotine, at a concentration of 1 mM, caused a statistically significant increase in micronucleus frequency following incubation for periods from 24 to 72 hours, while no change was detected in cell growth under the same conditions. Additionally, preincubation of human gingival fibroblasts with 1 mM nicotine strongly attenuated staurosporine-induced apoptosis. Cultures exposed to nicotine also exhibited an increase in reactive oxygen species, as determined by increased levels of 2,7-dichlorofluorescein. When cells were prelabeled with N-acetyl-cysteine, a substrate for glutathione synthesis, and catalase, an oxygen free radical scavenger, a significant reduction in cytogenetic damage was observed. The investigators concluded that the results of this study indicate that nicotine causes DNA damage, that it affects the activation of the apoptotic pathway and that it induces oxidative stress that which contributes to DNA injury in vitro.14
In contrast to the two in vitro studies reported above, another recently published study reported a nonmutagenic result for nicotine. In this study, nicotine was evaluated in the conventional in vivo mouse bone marrow assay following administration of single oral doses of 1 or 2 mg/kg nicotine to male mice. Bone marrow samples were taken and evaluated at 24 hours post-treatment from animals administered 1.0 mg/kg nicotine and at 6, 12 and 18 hours post-treatment from animals administered 2.0 mg/kg nicotine. The results of this study demonstrated that nicotine did not increase the micronucleus frequency in polychromatic erythrocytes, while the positive control, mitomycin C, yielded the expected results.

ГЕНОТОКСИЧНОСТЬ
Для изучения мутагенного действия различных концентраций никотина были использованы разнообразные тесты определения генотоксичности in vitro и in vivo. Как показали результаты исследований, некоторые проведенные тесты не подтвердили мутагенного действия никотина. Однако результаты других демонстрировали наличие некоторых признаков хромосомных повреждений.
Более углубленный анализ ранее проведенных исследований мутагенного действия никотина и его метаболитов представлен в заявке на регистрацию препарата NiQuitin CQ в виде трансдермального пластыря (раздел «Токсико-фармакологическая документация», Часть IIID).
Со времени подачи заявки на регистрацию NiQuitin CQ в форме трансдермального пластыря в научной литературе были опубликованы результаты еще трех исследований потенциального мутагенного и генотоксического действия никотина.

В другом исследовании, результаты которого были опубликованы недавно, изучалось повреждающее действие никотина на индукцию ДНК и апоптоз фибробластов десны человека с использованием in vitro блокирующего цитокинез микроядерного теста.
Результаты исследования показали, что действие никотина на фибробласты десны человека в концентрации 1 мг/мл, привело к статистически значимому увеличению числа микроядерных клеток через 24-72 часов после инкубации, вместе с тем изменений клеточного роста при этих же условиях зарегистрировано не было.
Кроме того, обработка фибробластов десны человека перед инкубацией раствором никотина в концентрации 1 мк/мг значительно ослабила процесс стауроспорин-индуцируемого апоптоза. В культурах, подвергшихся действию никотина, увеличилось число активных форм кислорода, о чем свидетельствовало увеличение содержания 2,7-дихлорофлюоресцеина.
Анализируя результаты исследования, ученые сделали вывод о том, что никотин вызывает повреждение ДНК - он влияет на активизацию апоптоза и нарушает процессы окисления, что приводит к повреждению ДНК in vitro.
В отличие от этих двух исследований in vitro, о которых сообщалось выше, в другой недавно опубликованной научной статье приводились данные об отсутствии мутагенного действия никотина. В этом исследовании эксперимент проводился в обычных условиях in vivo на мышах. Исследовался костный мозг самцов мышей после получения ими внутрь одной дозы никотина 1 или 2 мг / кг веса. Осуществлялся забор образцов костного мозга и их анализ через 24 часа после ведения животным дозы никотина 1,0 мг / кг веса и через 6, 12 и 18 часов после ведения дозы 2,0 мг / кг веса. The medicinal product Hepatil contains ornithine aspartate. It belongs to hepatoprotective substances. The mechanism of action of ornithine aspartate is a resultant of effects of two aminoacids being its components: ornithine and aspartic acid, which are characterized by detoxifying effects - binding the ammonia in two metabolic pathways, i.e. synthesis of urea and glutamine in the liver. They therefore accelerate the ammonia detoxification processes of the body. Both are substrates in the urea cycle and both stimulate the conversion of ammonia to urea. The liver is the place where final detoxification of ammonia takes place. Ornithine aspartate is more effective in the urea cycle than any one of the amino acids used separately. Initial detoxification is observed in the muscles and in the brain. In these tissues ornithine and aspartic acid are substrates in glutamate synthesis. It is a product of transamination reaction, being transposition of the amine group from a-amino acid to ketoglutarate. Glutamate binds ammonia leading to synthesis of non-toxic glutamine. Approximately L amount of the body ammonia can be inactivated in this pathway. The amine group of glutamate participates in the synthesis of carbamoyl phosphate which initiates the ornithine cycle. This reaction is catalysed by carbamoyl phosphate synthetase. In addition, aspartic acid by effect on the synthesis of nucleic acids favours the liver regeneration processes. In the condensation reaction of carbamoyl phosphate with aspartic acid, the product is carbamoyl aspartate, being a precursor in the pyrimidine nucleotides synthesis. Aspartic acid after transformation to oxaloacetate participates in the Krebs cycle, which leads to ATPsynthesis, thanks to which it may be indicated in conditions of inanition or after severe physical exercise. | Translation - Russian
Медицинский продукт Гепатил содержит орнитина аспартат. Является гепатопротектором. Механизм действия орнитина аспартата обусловлен действием двух аминокислот, из которых он состоит: орнитиновой и аспарагиновой кислот, которые обладают детоксицирующим действием – а именно, связывают аммиак в двух метаболических реакциях, синтезе мочевины и глютамата в печени. Поэтому они ускоряют процессы детоксикации аммиака в организме. Обе аминокислоты участвуют в синтезе мочевины и стимулируют превращение аммиака в мочевину. Это продукт реакции трансаминации, результат транспозиции аминовой группы от а-аминокислоты к кетоглумарату. Глутамат связывает аммиак, что приводит к синтезу нетоксичного глютамина.
Таким образом может быть инактивировано приблизительно L количество всего аммиака. находящегося в организме.
Аминовая группа глютамата участвует в синтезе карбамоил фосфата, который запускает орнитиновый цикл. Катализатором этой реакции является карбамоил фосфат синтетаза. Кроме того, аспарагиновая кислота за счет действия на синтез нуклеиновых кислот способствует процессам регенерации печени. Gay and Lesbian Studies in Brazil: A Field in Construction Traditionally, sexuality studies in Brazil have been related to a normalizing approach. The eye of the specialist has been trained to observe behaviours that did not conform with the imperatives of heterosexuality. The polarized, hierarchical worlds of the genders were unified and had a meaning within the framework of the idea of the complementary sexes. Therefore, heterosexuality would be the rule through which mankind would find the meaning of its own existence and reproduction.In recent years, this hegemonic view became the target of an intense process of deconstruction. Both debates and research on sexualities emerged outside the framework of medicalization of behaviors or the pathologization of non-heterosexual identities at Brazilian universities. Sexualities stopped being considered a naturalized place intended for reproduction, to be seen as a power-saturated locus. This deconstruction process is well in progress. This new approach in research first appeared in the area of social science, particularly over the last 30 years. The emergence of the HIV/AIDS epidemic and the works of Michel Foucault represent an important frame to outline a new look at sexual practices.Some of the priority issues in research studies are the critical thinking on issues such as homosexual marriage, sex reassignment surgeries, surveys on aggressions and murders of transvestites, lesbians, transsexuals and gays; the impacts of AIDS on sexual imaginary, the production and reproduction of homophobia, research on medical, religious and legal discourses, pornography and sexual education. Gradually, the discourses of social scientists have contributed significantly to the recognition of the historical development and the social character of sexual behaviors, and at the same time they have structured interpretations on the configuration of Brazilian sexual cultures. | Translation - Russian

Этот деконструктивный процесс идет полным ходом. Это новое направление в научных исследованиях впервые возникло в эру социальной науки и получило развитие в последние 30 лет. Moxidectin is currently being evaluated at the Onchocerciasis Chemotherapy Research Centre (OCRC) in Hohoe, Ghana in a phase 2, proof-of-concept study (protocol 3110A1-200-GH, OCRC 33). This study is ongoing, however, enrollment has been completed as of 30 June 2008, with a total of 172 subjects.The study is a randomized, ivermectin-controlled, double-blind, single ascending dose, parallel design, inpatient/outpatient study of 3 dose levels of moxidectin (2 mg, 4 mg, and 8 mg) in adult male and female subjects with different degrees of severity of O. volvulus infection. The primary objective of this study is to determine the safety and tolerability of orally administered moxidectin in adult subjects with O. volvulus infection as measured by the incidence of adverse events (AEs) and clinically significant laboratory test results. A secondary objective is to determine doses that are microfilaricidal and, more important, doses that are macrofilaricidal or macrofilaria-sterilizing. В настоящее время 2 фаза Исследования, подтверждающего концепцию использования моксидектина (протокол 3110A1-200-GH, OCRC 33), проводится в Научном центре химиотерапии онхоцеркоза (OCRC) в Hohoe, Гана (Onchocerciasis Chemotherapy Research Centre (OCRC), Hohoe), Исследование еще не завершено, однако набор субъектов для его проведения (всего 172) был завершен до 30 июня 2008г.Это рандомизированное, ивермектин-контрлируемое, двойное-слепое исследование применения трех однкратных доз моксидектина (2 мг, 4 мг и 8 мг), которое проводится параллельно в двух группах (стационарно/амбулаторно) субъектов мужского и женского пола с различной степенью тяжести инфекции, вызванной O. volvulus.Основная цель этого исследования – определение безопасности и переносимости моксидектина при приеме внутрь взрослыми субъектами, инфицированными O.
