Several recent observations suggest that Schwann cell (SC) differentiation, including myelin formation, is dependent upon the development of basal lamina which characteristically surrounds each axon-SC unit in peripheral nerve. This dependence can be tested in a neuron-SC culture system developed in our laboratory in which SC differentiation, including basal lamina formation and myelination, is faithfully reproduced. The use of serum-free, defined medium (DM) with this culture system allows axon-driven SC proliferation but not basal lamina formation or myelination. We previously demonstrated that ascorbic acid, in the presence of a nondialyzable serum factor(s), stimulates basal lamina assembly and myelin formation with similar dose- response relationships (Eldridge et al., 1987). We hypothesized that ascorbic acid acts to promote SC myelination indirectly, by enabling the assembly of basal lamina. We now provide support for this hypothesis by demonstrating the following.

(1) Pepsin-resistant triple- helical collagen molecules were produced only by SCs grown in the presence of ascorbic acid, suggesting that triple-helical type IV collagen may mediate the effect of ascorbic acid on basal lamina formation.

(2) The formation of myelin by oligodendrocytes, which myelinate axons in the CNS without the concomitant deposition of basal lamina, was little affected by ascorbic acid, suggesting that the biosynthesis and assembly of myelin per se does not require ascorbic acid.

(3) The provision of exogenous basal lamina matrix to SCs grown with neurons in DM without ascorbic acid promoted control levels of myelination (and basal lamina formation); the provision of exogenous fibrillar collagen matrix did not.

(4) Purified laminin promoted control levels of myelination in the absence of ascorbic acid, but purified type IV collagen and heparan sulfate proteoglycan (HSPG) did not. Laminin caused SCs to assemble basal lamina-like structures that contained not only laminin but also HSPG and non-triple-helical type IV collagen. Thus, several types of experiments demonstrate that SC myelin formation can be controlled by regulating the ability of the SC to assemble basal lamina, illustrating that acquisition of basal lamina is a crucial prefatory step for further SC differentiation.

최근 밝혀진 사실로는 미엘린 형성을 포함한 슈반 세포의 분화가 말초신경에서의 액손-슈반 세포 유닛 각각을 특징적으로 둘러싼 바닥판의 발달에 달려있다는 것이 있다. 이 의존성은 바닥판 형성과 신경세포 수초화를 포함한 슈반 세포 분화를 충실히 재현하도록 우리 연구실에서 개발한 뉴런-슈반 세포 배양 시스템에서 시험할 수 있었다. 이 배양 시스템에서 장액이 없는 제한 배지 (DM) 를 이용하여 액손에 의해 촉진된 슈반 세포를 분열시켰으나 바닥판 형성이나 신경세포 수초화는 그렇지 못했다. 우리는 이전에 투석 불가능한 혈청 내 요소(Serum factor)의 존재 하에서 아스코르빈산이 바닥판 정렬과 미엘린 형성을 촉진한다는 것을 확인했다. (엘드리지 연구팀, 1987) 우리는 아스코르빈산이 바닥판의 정렬을 가능하게 함으로써 슈반 세포의 수초화를 간접적으로 촉진한다는 가정을 세웠다. 이제 다음과 같은 것을 수행함으로써 이 가설의 증거를 제공할 것이다.

1) 펩신 저항성 삼중나선 콜라겐 분자는 아스코르빈산의 존재 하에서 성장한 슈반 세포에서만 생산되며, 이것은 삼중나선 4번 유형 콜라겐은 아스코르빈산이 바닥판 형성에 영향을 미치는 것을 매개한다는 것을 시사한다.

2) 중앙 신경계에서 바닥판의 축적을 수반하지 않은 채 액슨에 미엘린을 씌우게 되는 희소돌기아교세포(Oligodendrocyte)에 의한 미엘린의 형성은 아스코르빈산에 그다지 영향을 받지 않았으며, 이는 미엘린의 정렬과 생합성 그 자체는 아스코르빈산을 요구하지 않음을 의미한다.

3) 외생성(Exogenous) 바닥판 세포간질을 아스코르빈산이 없는 제한 배지의 뉴런에서 생장한 슈반 세포로 넣을 경우 수초화 (와 바닥판 형성) 의 컨트롤 등급이 상향되었다. 그러나 외생성 섬유질 콜라겐 매트릭스를 넣으면 그렇지 않았다.

4) 정화된 라미닌은 아스코르빈산이 없을 경우 수초화의 컨트롤 등급을 상향시켰으나, 정화된 4번 유형 콜라겐과 황산염 헤파란 단백당 (heparan sulfate proteoglycan, HSPG) 은 그렇지 않았다. 라미닌은 슈반 세포가 라미닌 외에 HSPG와 비삼중나선 Type IV 콜라겐 또한 포함하는 바닥판과 유사한 구조를 형성하도록 한다. 따라서 이러한 실험들로 인하여 우리는 슈반 세포에서의 미엘린 형성은 슈반 세포가 바닥판을 정렬하는 능력을 규제함으로써 조절할 수 있으며, 바닥판의 획득이 슈반 세포 분화가 진행되기 위한 가장 중요한 단계라는 사실을 알 수 있다.
