EXAMPLE 10: Transformation of L-Alanine into D-Alanine
[0049] Using the compounds of the present invention, L-alanine was converted into D-alanine as analyzed by 1H NMR spectra of FIG. 3.
[0050] In FIG. 3, an imine compound resulting from the reaction of an S-binaphthol derivative of Compound 5 with L-alanine in the presence of 3 equivalents of Et3N was 1H NMR-analyzed immediately after the reaction (a), 2 hours after the reaction (b), 3 hours after the reaction (c) and 48 hours after the reaction (d). As seen in these spectra, new peaks appear just after the reaction, which coincide with those of the imine formed between Compound 5 and D-alanine. 48 hours after the reaction, the peaks detected in D-alanine predominated over those detected in L-alanine, indicating that L-alanine was transformed into D-alanine. As known from the spectra of FIG. 3, 90% of L-alanine was converted into D-alanine through a reaction between Compound 5 and L-alanine after 48 hours. In contrast, transformation from D-alanine
to L-alanine seldom occurred. The reason for this is because Compound 5 is an S-binaphthol derivative. On the other hand, therefore, D-amino acid can be converted into L-amino acid using an R-binaphthol dernrative. In consequence, the compounds of the present invention can be used to obtain optically pure D-amino acid or L-amino acid from racemic amino acid. | Translation - Dutch

[0050] In FIG. 3 werd een imineverbinding, gevormd door reactie van een S-binaftolderivaat van Verbinding 5 met L-alanine in de aanwezigheid van 3 equivalenten Et3N, onmiddellijk na de reactie (a), 2 uur na de reactie (b), 3 uur na de reactie (c) en 48 uur na de reactie (d) geanalyseerd met behulp van 1H-NMR. Zoals in deze spectra te zien is, verschijnen er vlak na de reactie nieuwe pieken, die overeenkomen met die van het imine dat gevormd is tussen Verbinding 5 en D-alanine. 48 Uur na de reactie overtroffen de bij D-alanine waargenomen pieken degene die bij L-alanine waargenomen zijn, hetgeen aangeeft dat L-alanine in D-alanine omgezet is. Zoals bepaald uit de spectra van FIG. 3 werd 90% van het L-alanine in D-alanine omgezet door middel van een reactie tussen Verbinding 5 en L-alanine na 48 uur. De omzetting van D-alanine naar L-alanine kwam daarentegen zelden voor. De reden hiervoor is dat Verbinding 5 een S-binaftolderivaat is. Anderzijds kan daarom een D-aminozuren in het L-aminozuur worden omgezet met behulp van een R-naftolderivaat. During clinical trials, the following side effects have been observed. Their frequencies have been estimated as Common: affects 1 to 10 users in 100.
• headache
• sweating
• myalgia, arthralgia
• fever, generally feeling unwell (malaise), shivering, fatigue
• local reactions: redness, swelling, pain, ecchymosis, induration around the area where the vaccine is injected.
These reactions usually disappear within 1-2 days without treatment. Next to the above common side effects, the following side effects occurred after the vaccine came on the market:
• allergic reactions:
o leading to medical emergency with a failure of the circulatory system to maintain adequate blood flow to the different organs (shock) in rare cases,
o swelling most apparent in the head and neck, including the face, lips, tongue, throat or any other part of the body (angioedema) in very rare cases
• skin reactions that may spread throughout the body including itchiness of the skin (pruritus, urticaria), rash
• blood vessel inflammation which may result in skin rashes (vasculitis) and in very rare cases in temporary kidney problems
• temporary reduction in the number of certain types of particles in the blood called platelets; a low number of these can result in excessive bruising or bleeding (transient thrombocytopenia); temporary swelling of the glands in the neck, armpit or groin (transient lymphadenopathy) | Translation - Dutch
Tijdens klinisch onderzoek zijn de volgende bijwerkingen waargenomen. De frequenties ervan zijn als Algemeen ingeschat: betreft 1 tot 10 van de 100 gebruikers.
• hoofdpijn
• zweten
• myalgie, artralgie
• koorts, algemeen onwel voelen (malaise), rillingen, vermoeidheid
• lokale reacties: roodheid, zwelling, pijn, ecchymose, induratie rond de plaats waar het vaccin geïnjecteerd is.
Deze reacties verdwijnen gewoonlijk binnen 1-2 dagen zonder behandeling. Na het op de markt komen van het vaccin kwamen naast de bovengenoemde algemene bijwerkingen de volgende bijwerkingen voor:
• allergische reacties:
o met medisch noodgeval tot gevolg, waarbij er niet voldoende bloedtoevoer naar de verschillende organen gehandhaafd kon worden (shock) in zeldzame gevallen,
o zwelling, meest duidelijk in het hoofd en de hals, waaronder het gezicht, de lippen, tong, keel of elk ander deel van het lichaam (angio-oedeem) in zeer zeldzame gevallen
• huidreacties die zich over het lichaam kunnen verspreiden, waaronder jeuk aan de huid (pruritus, urticaria), huiduitslag
• bloedvatontsteking die kan leiden tot huiduitslag (vasculitis) en in zeer zeldzame gevallen tot tijdelijke nierproblemen
• tijdelijke vermindering van het aantal van bepaalde deeltjes in het bloed (bloedplaatjes); een laag aantal hiervan kan leiden tot bovenmatige kneuzingen of bloedingen (voorbijgaande trombocytopenie); tijdelijke zwelling van de klieren in de hals, oksel of lies (voorbijgaande lymfadenopathie) |
