Could anyone please tell me whether as an English-Spanish translator, I'd have to take the paper in Spanish-English too?

It is a requirement? I think it is.

No, there's no need! In fact, as you normally translate into your mother tongue, most people in your position would only translate into Spanish. If you have a second language, say French, you could also choose to do the Diptrans from French into Spanish, but that would be a completely different exam and therefore qualification. The Diptrans consists of 3 papers : general translation (3 hours), and 2 semi-specialized papers, each lasting 2 hours. Have a look at the Iol website - you'll find all the info you need there. Who are you taking the diploma with?
For the IoL Diploma in Translation each language combination is separate so you would only have to do English to Spanish.
