Odontoss�ese tooth synthesis / Explanation: Research Symposium 1999 - Protease activity must be crucial for the structural rearrangements in the extracellular matrix necessary for bone and tooth synthesis. Hydroxyapatite is a main mineral component of bone and teeth and it is known that it affects protease activity in some ways. This project investigates the affects of both hydroxyapatite and the thiol protease inhibitor cystatin on the thiol protease papain. The protease activity was measured by the degradation of the dye-linked substrate protein azocasein. Papain activity was found to obey Michaelis-Menten kinetics with this substrate. Papain was shown to bind hydroxyapatite in solution, but the association was very weak. The results of the incubation with cystatin and hydroxyapatite were inconclusive.
