Em ruminantes, o processo de vacinação ocorre predominantemente pela via subcutânea e com o auxílio de seringas, as quais são utilizadas para inúmeros animais. Tal procedimento, favorece a disseminação de doenças de veiculação hematogênica, o que coloca em risco as práticas de biossegurança adotadas em programas de controle de doenças infecciosas. Uma alternativa para contornar este problema é o desenvolvimento de vias alternativas de vacinação, a quais não utilizem objetos perfurocortantes (sharp). Neste contexto, o presente estudo objetivou desenvolver um dispositivo lipídico, contendo nanopartículas de quitosana capaz de carrear intacta uma proteína de até 5,7 KDa através da mucosa gastrintestinal de ruminantes. Para tanto, insulina cristalina humana (Novonordisk- Montes Claros, Brasil) foi encapsulada em nanopartículas por complexação espontânea entre chitosan (Aldrich – Wisconsin, USA) e o sodium tripolyphosphate (Dinâmica- Diadema, Brasil) e posterior incorporação mecânica em uma matriz lipidica saturada e fundida à 28°C.
