Procedure Instructions
A. Purification of PCR fragments using Promega Wizard Protocol
1. Load and run the agarose gel using a standard protocol and running conditions.
2. Weigh a 1.5-ml vial for each DNA fragment to be isolated and record the weight.
3. Visualize the gel under UV-lamp and the DNA fragment in a minimal volume of agarose using a clean scalpel. Transfer the gel slice to weighed pre-labeled vial and record its weight. Subtract the weight of the empty vial from the total weight to calculate the weight of the gel slice.
4. Add Membrane Binding Solution at a ratio of 10 ul of solution per 10 mg of agarose slice. (Since the slices may be different in size by 5-10%, add the same amount of Membrane Solution to each vial based on calculation made for the heaviest slice).
