Using this method, actively growing bacteria are exposed to the test article in suspension for a specified period (often 1 h), centrifuged, washed free of the test material, and plated out in top agar as per the standard method. At the end of the suspension exposure period, the number of viable surviving bacteria is quantified by diluting and plating them in complete medium with excess histidine/tryptophan. In the absence of any effect of the material on bacterial survival, mutagenicity will be evident as an absolute increase in revertant colonies in the normal way.
