A microtiter plate (spelled Microtiter is a registered trade name in the United States) or microplate or microwell plate or multiwell,[1] is a flat plate with multiple "wells" used as small test tubes. The microplate has become a standard tool in analytical research and clinical diagnostic testing laboratories. A very common usage is in the enzyme-linked immunosorbent assay (ELISA), the basis of most modern medical diagnostic testing in humans and animals.

The fluctuation test is performed by treating amino acid-requiring bacteria in suitable medium, either with or without a metabolizing system (S9 fraction or isolated hepatocytes), with the chemical to be tested and with a trace amount of the required amino acid (to allow a few generations of growth during which mutations to amino acid independence may be expressed).
This mixture is immediately dispensed in small aliquots into the wells of disposable plastic trays and incubated for 16-18 hours. Selective indicator medium is then added to each well so that only revertants can continue to grow. The bacteria release acid on growth to full turbidity; therefore, a pH indicator, bromocresol purple, is added. At full turbidity the pH changes from 7 (purple) to 5.2-6.8 (yellow). The yellow wells are those in which one or more reversions to amino acid independence have occurred. The test has been modified since the original papers by Green et al. (1976, 1977a) to use disposable plastic trays, which avoid the problems of decontaminating and washing glass tubes and occupy less incubator space.
