“Se ven, hablan y actúan de manera distinta a la nuestra”

I would favor the preposition “de”, in the sense of their having a different appearance, speaking and behaving in a manner different to the one we would or do. Now, if instead of “differently from us”, your text read “different to us”, then I would favor the preposition “a”, “de manera distinta a la nuestra”.

also can be translated as DESDE, but not in this context.-
The full sentence is: "Ellos lucen, hablan y actúan diferente de/a nosotros.-
