Hi Many, here you have some option:

-Me entiendes? : This one is the standard translation. Polite, exact, no ambiguities.

-Entendido? : Use this right after you have given an ORDER or explained something important... "Take this book and study the whole book, do you understand me?!" .... "toma este libro y estúdiatelo entero, ¿ENTENDIDO?". It is a bit rude :)

-Lo pillas/captas? : the ironical version, when you really are spoiled and do not want to repeat the whole thing...someone may think it's a bit rude, but it's funny, in a way. "The green book is MINE... do you understand me?" "El libro verde es MÍO... lo captas? Lo pillas?"

Explanation:
"Do you" is referring to a person and the only way to do this in spanish is by starting the question this way "Usted" then "me entiende" which of course means "understand me". Also, remember that in spanish there is a backwards question mark at the beginning of the question and the usual one at the end.
