Source text - English The AtSEC61 subunit protein was incubated for various times in the SGF, and the remaining protein was subjected to electrophoresis and visualized by western blot analysis. The E. coli-produced AtSEC61 subunit protein was rapidly degraded in SGF (Figure 3A). AtSEC61 subunit protein (molecular weight ca. 7,000, the most intensely staining band in this size range) is readily detected in the time 0 sample and in the G-con samples. However, no full-length AtSEC61 subunit protein is visible in the sample removed at 0.5 min after the initiation of the reaction. Some immunoreactive bands representing contaminating proteins of varying molecular weights (6000, 10,000, 25,000, 36,000,) were visible at all time points. Other immunoreactive protein bands at 20,000, 32,000, 63,000 and 98,000 molecular weights were also rapidly degraded similar to the AtSEC61 subunit protein. The results shown in the blot pictured in Figure 3B in which the immunoaffinity-purified rabbit anti-SEC61 peptides was used, confirm the identity of the AtSec61 band and its rapid digestibility.
Results of the current study show that the AtSEC61subunit protein is highly digestible under the simulated digestion conditions, which is typical of most dietary proteins exposed to the proteases of the mammalian digestive tract. This result demonstrates that the AtSEC61 subunit protein does not share one of the characteristics of some food allergens, resistance to digestive degradation. Furthermore, this result was as expected since the AtSEC61 subunit protein is ubiquitous and highly conserved in eukaryotes, and is present in many plant species with a history of safe food use.
Collectively, results of the above studies demonstrate that in the unlikely event the AtSEC61 subunit protein is expressed at extremely low levels (below the limit of detection of the western blot assay) in CV127 tissues, the AtSEC61 subunit protein does not posses the attributes of known protein food allergens, is not homologous to known protein toxins, and hence poses no risks for human and animal consumption.
Translation - Portuguese A subunidade da proteína AtSEC61foi incubada em SGF por vários intervalos de tempo diferentes, e o restante da proteína foi submetida a uma eletroforese e visualizada por análise de Western Blot. A subunidade da proteína AtSEC61 produzida por E. coli foi rapidamente degradada no SGF (Figura 3A). A subunidade da proteína AtSEC61 (peso molecular ca. 7000, a banda mais intensamente marcada nessa faixa de tamanho) é prontamente detectada na amostra de tempo 0 e nas amostras G-con. Entretanto, o comprimento total da subunidade da proteína AtSEC61 não é visível na amostra removida em 0,5 minutos após o início da reação. Algumas bandas imunorreativas representando proteínas contaminantes de diversos pesos moleculares (6.000, 10.000, 25.000, 36.000) foram visíveis em todos os intervalos de tempo do ensaio. Outras bandas de proteínas imunorreativas nos pesos moleculares 20.000, 32.000, 63.000 e 98.000 também foram rapidamente degradadas assim como a subunidade da proteína AtSEC61. Resultados do presente estudo mostram que a subunidade da proteína AtSEC61 é altamente digerível sob as condições de digestão simuladas, que é típica da maioria das proteínas da dieta expostas a proteases do trato digestivo dos mamíferos. Esse resultado demonstra que a subunidade da proteína AtSEC61 não compartilha uma das características de alguns alérgenos alimentares, resistência à degradação digestiva. Além do mais, esse resultado foi como o esperado, uma vez que a subunidade da proteína AtSEC61 é ubíqua e altamente conservada em eucariotos, e também é presente em muitas espécies de plantas com histórico de alimentos seguros para consumo.
Coletivamente, resultados dos estudos acima demonstram que no improvável evento da subunidade da proteína AtSEC61 estar expressa em níveis extremamente baixos (abaixo do limite de detecção do ensaio de Western Blot) em tecidos CV127, a subunidade da proteína AtSEC61 não possui os atributos de proteínas conhecidas como alimentos alergênicos, não é homóloga a toxinas protéicas conhecidas, e por isso não apresenta risco para consumo humano e animal.
