Die jeweils aktuelle Standardkodierung, Haltbarkeit, Konzentration und Aliquotierung ist der aktuellen Anlage 5 zur QA009 zu entnehmen.
Standardvorbereitung:
Am Tag der Analytik werden zwei Standardvials für ca. 0,5 h bei RT aufgetaut und mit der Pipette homogenisiert.
Es wird 3 Mal gemischt und eine Blasenbildung ist zu vermeiden.
Der Standard wird in ein HPLC-Vial pipettiert und das HPLC-Vial verschlossen.
Am Beginn jeder Probensequenz (siehe 10.3.3.2) erfolgen 2 Standardinjektionen zur Equilibrierung, im Anschluß daran nochmals 3 Standardinjektionen, die zur SST Auswertung herangezogen werden.
Spätestens nach jeweils 10 Probeinjektionen erfolgen wiederum 3 Standardinjektionen, die ebenfalls zur SST Bewertung herangezogen werden.
Bei den Proben handelt es sich um Drug Product, Drug Substance und Prozessintermediate des MOD-4023 Herstellungsprozesses.
Die Lagerung der Proben erfolgt gemäß den Vorgaben im LIMS bzw. des Prüfprotokolls

Freigabe bei 5 ± 3°C, Stabilitäten gelten LIMS Vorgaben
DP Stabilitätsproben (-20 ± 5 °C): bei -20 ± 5 °C
Säulenauftrag:
Um einen Säulenauftrag von 20 µg zu erhalten wird das Injektionsvolumen entsprechend angepasst (Bereich: von 5 – 100 µL).
Es wird 3 Mal gemischt und eine Blasenbildung ist zu vermeiden.
Es wird zum Mischen der Probe und zur Probenverdünnung jeweils eine extra Pipettenspitze verwendet.
Es wird das entsprechende Injektionsvolumen + mind. 30 µL Zuschlag abgefüllt und das HPLC-Vial verschlossen.
Einfachbestimmung (einfache Probenaufarbeitung, einfache Injektion)
