Source text - Portuguese O HIV provoca uma resposta imune inflamatória que induz a exaustão das células T CD4+ e evoluem para sequestrar os mecanismos de vesiculação celular regulada de citocinas e moléculas de sinalização. Tornou-se evidente que o HIV não só usa a via de vesiculação para sua formação e disseminação, mas também explora a transferência de vesículas extracelulares das junções imunes. As vesículas extracelulares expressam diferentes marcadores em sua superfície dependendo da sua célula de origem que podem conter RNA, DNA e proteínas, e fornecem informações valiosas sobre o estado de saúde da célula, entre elas, as células formadoras do osso. Desta maneira, iremos trabalhar com a interação do HIV e metabolismo ósseo no envolvimento de vesículas extracelulares e anexina V para melhor compreensão dos mecanismos moleculares do HIV e células ósseas. O objetivo do estudo será avaliar a concentrações de vesículas extracelulares e a expressão de anexina V sobre a proliferação e diferenciação das células ósseas em indivíduos controladores de elite, indivíduos suprimidos em uso de TARV e não-controladores.

HIV causes an inflammatory immune response that induces the exhaustion of CD4+ T cells and evolves to sequester the mechanisms of regulated cellular vesiculation of cytokines and signaling molecules. It is now evident that not only does HIV use the vesiculation pathway for its assembly and dissemination, but also exploits the transfer of immune junction extracellular vesicles. Extracellular vesicles express different markers on their surface, depending on the cell where they originate from. They may contain RNA, DNA and proteins, and provide valuable information about the cell’s health status, including bone-forming cells. Therefore, we will work on the HIV and bone metabolism interaction on the involvement of extracellular vesicles and annexin V to gain a better understanding of the molecular mechanisms of HIV and bone cells. The goal of this study will be to assess the impact of extracellular vesicle concentration and annexin V expression on proliferation and differentiation of bone cells in elite controller individuals, suppressed individuals under ARVT and non-controllers.
