English to Malay: Research on new neuraminidase inhibitors
Source text - English With phenomenal impact of the possibility of flu pandemic, and the present threat of current antiviral neuraminidase inhibitors (oseltamivir and Zanamavir), the search for new anti-influenza chemotheraputic is currently in dire need. This project thus aimed at discovering novel neuraminidase inhbitors employing structure based drug design approaching from natural product, chemical synthesis and computer aided drug design. The project was strengthened with the development of biological assays for inhibition of nueraminidase and viral replication (subproject 4&5). Natural product subgroup (subproject 2) have found few spots in TLC demonstrating the existence of few single compounds in tamarind and mango. 3 structures were identified, i.e. beta-sitosterol, stigmasterol and a new compound, 6,6’-oxybis(4chloromethyl)picolinaldehyde for tamarind, while for mango they isolated and identified 3-Hydroxy-3-(Methoxycarbonyl) pentanedioic acid, 1,5-Dimethylcitrate monohydrate, 2-Hydroxybenzoic acid. Although, bioassay confirmed activity of the extracts and fractions of the plants, the compounds isolated demonstrated insignificant activity. The synthesis group (subproject 3) synthesised 24 novel benzoic acid derivatives of which was designed intuitively to target nueraminidase and one compound is active against the bacterial neuraminidase. The computer modelling group (subproject 1) who screened Malaysian plants demonstrated that they could pick up 7 plants with confirm bacterial and neuraminidase activity, in addition to tamarind and mango. They further carried out pharmacophore design to derivatise natural products and vanillin derivatives designed proved to be higly active. In fact one compound, VEPH, demonstrated activity better than DANA, tamiflu the current leading inhibitor of the enzyme. Due to restrictive of budget, in April 2010 this project has to be on hold as no additional fund is given to proceed with the bioactivity studies. However, as we received funding from MOSTI we managed to carry out viral replication studies and toxicities of these compounds. This compound along with 3 other derivatives also demonstrated superior toxicity profile on cell culture. VEPH is also shown to be able to inhibit the viral replication of H5N1.
Translation - Malay Berdasarkan kesan fenomena pandemik jangkitan flu (selesema) yang mungkin terjadi dan ancaman-ancaman semasa terhadap tahap keberkesanan perencat-perencat antivirus neuraminidase (oseltamivir dan zanamivir), pencarian agen kemoterapi anti-influenza yang baru sememangnya amat diperlukan. Oleh hal yang demikian, projek ini mensasarkan untuk menemui perencat-perencat enzim neuraminidase yang baru dengan menggunakan rekabentuk drug berasaskan struktur dengan menumpukan kepada bahan-bahan daripada sumber alam, sintesis kimia dan rekabentuk drug menggunakan komputer. Projek ini telah diperkuatkan lagi dengan pembangunan asai biologi untuk perencatan enzim neuraminidase dan replikasi virus (Sub-projek 4 dan 5). Sub-kumpulan natural product (Sub-projek 2) telah berjaya menemui beberapa titik tompokan pada TLC (Kromatografi Lapisan Nipis) yang menunjukkan kewujudan beberapa sebatian tunggal dalam asam jawa dan mangga. 3 struktur telah dikenalpasti, iaitu beta-sitosterol, stigmasterol dan sebatian baru, 6,6’-oksibis(4-klorometil)pikolinaldehid daripada asam jawa, manakala daripada buah mangga, mereka telah berjaya mengasingkan dan mengenalpasti asid 3-Hydroksi-3-(Metoksikarbonil) pentanadioik, 1,5-Dimetilsitrat monohidrat dan asid 2-Hidroksibenzoik. Walaupun asai biologi telah membuktikan bahawa terdapat aktiviti daripada ekstrak dan pecahan-pecahan ekstrak daripada pokok-pokok tersebut, sebatian tunggal yang telah diasingkan bagaimanapun menunjukkan aktiviti yang tidak signifikan. Kumpulan sintesis (sub-projek 3) telah mensintesis 24 sebatian terbitan asid benzoik yang baru, yang mana telah disintesiskan berdasarkan jangkaan dan andaian pada keupayaan sebatian tersebut untuk merencatkan enzim neuraminidase, dan satu daripada sebatian-sebatian tersebut adalah aktif terhadap neuraminidase bakteria. Kumpulan pemodelan komputer (sub-projek 1) yang telah menganalisa tumbuhan-tumbuhan dari Malaysia telah menunjukkan bahawa 7 tumbuhan mempunyai aktiviti terhadap bakteria dan neuraminidase, selain asam jawa dan mangga. Mereka seterusnya telah melakukan rekabentuk farmakofor terhadap sebatian-sebatian daripada sumber alam ini dan pecahan-pecahan vanillin yang telah terbukti sebagai sangat aktif. Malah satu sebatian, VEPH telah menunjukkan aktiviti yang lebih baik daripada DANA dan Tamiflu® yang merupakan perencat enzim yang paling aktif pada masa kini. Disebabkan terdapat kekangan kewangan, projek ini terpaksa ditangguhkan pada April 2010 disebabkan tiada dana tambahan diberikan untuk meneruskan kerja-kerja penyelidikan berkenaan bioaktiviti. Walau bagaimanapun, dana daripada MOSTI telah membolehkan kami melakukan penyelidikan terhadap replikasi virus dan toksisiti sebatian-sebatian tersebut. Sebatian VEPH dan 3 sebatian terbitan lain telah menunjukkan ciri toksisiti yang tinggi terhadap kultur sel. VEPH juga telah menunjukkan keupayaan untuk merencatkan replikasi virus H5N1.
