Procedure for Plaque Immunoassay
Select PLAQUES of appropriate size and number
Select cell monolayers that have been infected for 4 to 8 days.
Check for PLAQUE size and number either by inverting a PLATE without the lid over a light box to visualize the PLAQUES or by holding a PLATE up to the light.
PLAQUES should be at least 1 mm in diameter.
Positive and negative controls must be included in each assay.
Prepare PLATES selected
PLATES with agarose overlay should be inverted and the agarose removed by flipping it out with a spatula.
Visually check for monolayer and PLAQUE retention.
Wash the remaining overlay from the monolayer by gently pipetting 15 - 30°C PBS-0.1% Ca/Mg onto the PLATES (use approximately 5 mL per 60 mm dish).
