Source text - Italian Abstract. Sporadic Alzheimer’s disease (AD) and Parkinson’s disease (PD) are two of the most common neurodegenerative
diseases and as such they represent major public health problems. Finding effective treatments for AD and PD represents an
unmet and elusive goal largely because these diseases are chronic and progressive, and have a complicated and ill-understood
pathogenesis. Although the underlying mechanisms are not fully understood, caffeine, the most commonly ingested psychoactive
drug in the world, has been shown in human and animal studies to be protective against AD and PD. One mechanism implicated in
the pathogenesis of AD and PD is blood-brain barrier (BBB) dysfunction and we reported recently that caffeine exerts protective
effects against AD and PD at least in part by keeping the BBB intact. The present review focuses on the role of BBB dysfunction
in the pathogenesis of AD and PD, caffeine’s protective effects against AD and PD, and potential mechanisms whereby caffeine
protects against BBB leakage. | Translation - English Abstract: L’Alzheimer (AD, dall’inglese Alzheimer’s disease) e il Parkinson (PD, dall’inglese Parkinson’s disease) sono due delle malattie neurodegenerative più comuni e in quanto tali rappresentano i problemi principali della salute. Trovare degli efficaci trattamenti per l’Alzheimenr ed il Parkinson è un obiettivo sfuggente e non ancora raggiunto principalmente perché queste malattie sono croniche e progressive, ed hanno una patogenesi complicata e poco compresa. Sebbene i meccanismi alla base non sono pienamente compresi, è stato dimostrato che la caffeina, la sostanza psicoattiva più comunemente ingerita in tutto il mondo, ha un effetto protettivo contro l’AD e il PD negli studi [eseguiti su] umani ed animali. Un meccanismo implicato nella patogenesi di AD e PD è il malfunzionamento [o anche disfunzione] della barriera ematoencefalica (BBB, dall’inglese blood-brain barrier) e recentemente abbiamo riportato che la caffeina esercita effetti protettivi contro l’AD e il PD, almeno in parte, mantenendo intatto la BBB.
