Source text - Chinese 生物信息学分析显示TMCO1与肺腺癌之间可能存在联系,但是目前没有研究明确TMCO1是否参与肺腺癌发生发展的过程,因此我们期望通过本次研究来初步明确TMCO1与肺腺癌的关系以及其可能的调控机制。免疫组化染色发现,肺腺癌组织中TMCO1的表达明显高与正常组织。随后我们在A549细胞中敲除TMCO1,MTT发现A549细胞增殖能力下降,并且免疫细胞化学染色显示,敲除TMCO1会下调Bcl-2的表达以及上调Caspase-3和Caspase-9蛋白。此外,敲除TMCO1导致A549细胞迁移能力受到抑制,MMP-2和MMP-9表达降低,同时N-cadherin和Vimentin的表达下调,而E-cadherin表达上调。通过本次研究,我们首次发现TMCO1与肺腺癌存在联系,并且可以通过抑制TMCO1的表达来对肺腺癌细胞的增殖和迁移过程起到负向调控作用,表明了TMCO1可用于临床治疗肺腺癌的潜在可能性。 | Translation - English Bioinformatics analysis showed that TMCO1 might be associated with lung adenocarcinoma, but no research determined whether TMCO1 is involved in the development of lung adenocarcinoma. Therefore, we expect to identify the relationship between TMCO1 and lung adenocarcinoma as well as their potential regulatory mechanisms through this research. First, we found that the expression of TMCO1 in lung adenocarcinoma tissues was significantly higher than that in normal tissues from Immunohistochemical staining. Then, we knocked out TMCO1 in A549 cells and discovered that the proliferation of A549 cells decreased by MTT assay. At the same time, immunocytochemical staining uncovered that knockout of TMCO1 could down-regulate Bcl-2 expression and up-regulate Caspase-3 and Caspase-9 proteins. In addition, knockout of TMCO1 resulted in the inhibition of A549 cells’ migration, reduction of MMP-2 and MMP-9 expression, down-regulation of N-cadherin and Vimentin, and up-regulation of E-cadherin expression. Based on this research, we discovered for the first time that TMCO1 is associated with lung adenocarcinoma. Specifically, inhibiting the expression of TMCO1 could negatively regulate the proliferation and migration of lung adenocarcinoma cells, which indicates a potential possibility of TMCO1 being used for clinical treatment of lung adenocarcinoma. |
