The "a" sound will be the "u" sound in the Enclish word "but"" short and clear. Though written "iisutaa" in ROMAJI, the "u" will not be pronounced. Should you need a sound, give me a yell. +64 9 832 5666 or Skype name "Yoohoo Fujisawa"

ハッピーイースター
ハッピーイースター Happii Iisutaa
pronounced virtually the same as in English
Easter is not very big in Japan yet, so probably very few people would know what you are talking about.

御復活おめでとうございます (gofukkatsu omedetou gozaimasu)
This means "happy resurrection" and is what people said on Easter at a Catholic church I went to in Japan. So if you're saying it in a religious context, you can use this phrase. But otherwise, use "happii iisutaa" as suggested by the other answerers.

イースター おめでとう
easter is "イースター" that's why it's "イースター おめでとう ".
Example sentence:
たんじょうび おめでとう
