I like Scott's answer a lot. You should definitely use Scott's answer if you are talking casually.

My answer is an idiomatic phrase that exactly means "Chance of a Lifetime". This works beautifully if you are writing a novel or an article, but is bit more scholarly/traditional. (I bet many Japanese youths don't even know this meaning... how sad.)

Senzai Ichiguu (千載一遇)is a so-called Yoji-jukugo (four-word-idiom), because it consists of 4 chinese characters (Kanji).
The first 2 characters, Senzai, means 100 years.
The last 2 characters, Ichiguu, means Meeting once.
Kouki is the same meaning as the one that was listed by Dr.Shmidt.

Dr. Scmidt's answer is fine. But if you are interested in some alternatives, including the Japanese saying that I suggest above, then please see below.

I give the literal translations first. In all alternatives the loan word "chansu" (meaning pretty much the same as chance in the phrase you wish to translate) can be replaced by "kouki" which means pretty much the same thing in Japanese (as opposed to a loan word)

A chance (good opportunity) that only happens once in a thousand years (Japanese saying)
zenzaiichigu no chansu (kouki)
千載一遇のチャンス[好機]

A chance (good opportunity) that won't come again.
isshou ni mata to nai chansu [kouki]
一生にまたとないチャンス[好機]

A once in a lifetime chance (good opportunity)
Isshou ni ichido no chansu [kouki]
