For the majority of classically autistic children without a known medical disorder, genetic factors seem of primary importance, with twin studies demonstrating a heritability of over 90%, almost certainly due to multiple interacting genes rather than a single major gene. Linkage studies spanning the entire genome have identified several susceptibility loci. Unlike most such claims in psychiatric genetics, these localisations have been independently replicated for at least two regions (on chromosomes 7q and 2q). The heritable phenotype seems to be broad, stretching from classical autism at the one extreme to mild partial variants at the other extreme. The recurrence rate in siblings is roughly 3% for narrowly defined autism, but is about 10-20% for milder variants.
