The probe was radiolabelled with a-32P-dCTP (3000 Ci/mmol, Amersham) by a random priming method. Hybridization was performed in 50% formamide, 5X SSC (0.75M NaCl, 0.075M sodium citrate), Denhardts solution (0.1% Ficoll, 0.1% polyvinylpyrrolidone, 0.l% BSA), and 0.l% SDS for 16 h at 42¡ãC.

SSC" is a abbreviation of sodium chloride and sodium citrate solution, which used to transfer nuclear acid from gel to membrane ( 20 *SSC or 10*SSC or 5*SSC with alkaline). In Roche's DIG system, they said that the high solt concentration SSC shows much efficiently than the others like valcuum or electrophoresis.

To XiaoRan: The real issue of importance here is the final concentrations of constituents in the assay. How you get there needs to be clarified ¨C 0.5 x of what, 2x of what, concentration of 5x (see above), especially when you decide to publish your data and communicate them to the others to understand and therefore better appreciate your work.

: yes. the explanation of SSC is very good. But solutions such as 0.5 X buffer can also be used as working buffer. 5X solution can be diluted 5 times into a working buffer (1X), but it can also be diluted 10 times to make a working buffer (0.5X)

Theoretically yes, but in conventional and our lab 1X is the final concentration that you obtain from 5X in this instance, otherwise you would call initial concentrate 10X, 12X etc. Naturally, read your recipe carefully when using for the first time

0.75M NaCl is not saline, that is why. Once 5x SSC is diluted to 1x then you get saline sodium citrate. Namely, saline (physiologic solution) is 0.15M NaCl, which is 300 milliosmolar that is isotonic

I myself had once choose a not-best answer, but after that I always think more than twice of a professional's suggestion....

not theoretically yes, but practically yes. I have done research work in this field for 7 years, and I use 0.5X solution as a working buffer for millions of times. In addtion, sometimes we also use 2X solution as a working buffer.
